Molecular Characteristics of Novel Mono-Reassortant G9P[8] Rotavirus A Strains Possessing the NSP4 Gene of the E2 Genotype Detected in Tokyo, Japan

Abstract: INTRODUCTION Rotavirus A (RVA) is a major cause of gastroenteritis in infants and young children worldwide. In 2016, RVA caused 128,500 deaths in children under 5 years of age globally (1). Although most of these deaths occurred in developing countries, RVA imposes a huge burden even in developed countries, including Japan (2-4). Live attenuated RVA vaccines were introduced in Japan in 2011 (Rotarix ® , GlaxoSmithKline Biologicals, Rixensart, Belgium) and in 2012 (RotaTeq ® , Merck & Co., Inc., Kenilworth, NJ,… Show more

“…It is notable that three G9P[8] strains are reassortants possessing DS-1 like NSP4 genes in the genetic background of Wa-like rotavirus ( Figure 7 , E2). The reassortant G9-P[8]-I1-R1-C1-M1-A1-N1-T1-E2-H1 were also detected in Jinzhou, China, and Japan [ 66 , 67 ]. Since the identification of NSP4 genotype is not included in the routine surveillance, it is not clear when this kind of reassortant appeared in Wuhan.…”

“…This strain has not been previously reported in South or North America, but has a highly similar identity to sequences detected between 2010 and 2016 in Asia [ 63 ]. In the same line of evidence, the G8 sequences found in Trebal also shared a 99% nucleotide identity with sequences detected in 2013 in Thailand [ 64 ] and between 2017 and 2018 in Japan [ 65 ].…”

“…In one associated paper, these closely related samples were reported as minor G2P [4] variants circulating in the Mie prefecture in 2017 [37]. However, this variant was also detected in Tokyo in 2017, where G2P [4] was the dominant genotype, accounting for 40% of samples [38]. The outbreak samples also consistently clustered with Hungarian G2P [4] from 2012, where this genotype accounted for 13.5% of the samples genotyped in 2012 [39].…”
