Molecular Biology of Glucocorticoid Signaling

Abstract: Well-defined as signaling hormones for the programming of cell type-specific and context-dependent gene expression signatures, glucocorticoids control experience-driven allostasis. One unifying model is that glucocorticoids help maintaining the integrity and plasticity of cellular networks in changing environments through the mobilization of cellular energy stores, profiling of gene expression, and changes in the electrical and morphological properties of cells. The nucleus is their primary site of action, yet… Show more

“…The effects of dose and duration recapitulated by the acute and chronic stress paradigms were discussed earlier. The context at exposure such as learning at the time of glucocorticoid oscillation peak could produce unique effects, temporally restricted, that are the results of the coupling of neuronal activity with glucocorticoid signaling in select neuronal networks [ 64 , 121 ]. For example, a single injection of glucocorticoids within minutes of motor skill training enhanced motor learning.…”

“…glucocorticoid response elements, or GREs) of target genes, and alter gene expression. GR target genes may comprise more than 10% of the genome and include those involved in metabolism, immunity, and cell survival and growth, among others 7 . For example, in white adipose tissue (WAT) of terrestrial mammals, GCs increase fatty acid availability and adipocyte differentiation by upregulating expression of lipases and pro-adipogenesis factors (e.g.…”

“…Glucocorticoids (GCs) are essential for survival and serve a major role in embryonic development, tissue homeostasis and in the regulation of the inflammatory response ( 1 , 2 ). In breast, the functions of GCs are complex and depend, in part, if they are linked to their receptor (GR) and consist of the control of milk secretion, differentiation and apoptosis ( 3 ).…”
