The acceptability of potatoes for processing chips and French fries is largely dependent on the color of the finished product. Most potato cultivars and varieties stored at temperatures below 9-10°C are subjected to low temperature sweetening (LTS) which result in the production of bitter-tasting, dark colored chips and French fries which are unacceptable to consumers. However, storing tubers at low temperatures (i.e., <10°C) has many advantages such as lowered weight loss during storage, natural control of sprouting, and reduction/elimination of chemical sprout inhibitors. Our earlier research results on LTS suggested a role for pyruvate decarboxylase (PDC) in LTS-tolerance. In the present study, the role of PDC was examined whereby the potato variety Snowden was transformed with Arabidopsis cold-inducible pyruvate decarboxylase gene 1 (AtPDC1) under the control of promoter rd29A. Two transgenic plants were selected and storage studies were conducted on tubers harvested from one of the transgenic lines grown under green house conditions. Transgenic tubers showed higher Agtron chip color score indicating lighter chip and lower reducing sugar and sucrose concentrations compared to the untransformed tubers during the storage periods studied at 12°C and 5°C. These results suggest that overexpression of pyruvate decarboxylase gene resulted in low temperature sweetening tolerance in the transgenic Snowden.

Waterlogging is a major factor limiting barley grain yield worldwide. Climate change will likely increase this water stress in Northern Europe. Breeding for waterlogging tolerance (WLT), as for other abiotic stresses, is difficult, but identification of genetic markers linked to genes affecting WLT could facilitate the breeding process. To identify a suitable marker population, parents of 14 double-haploid (DH) barley populations were tested for segregation of biomass growth reduction in waterlogged soil. The most interesting was found in the offspring from crossing cv. Psaknon and breeding line (SLUdt1398 9 Mona 4 ). Hence, 120 DH-lines derived from this cross were phenotyped for the chlorophyll fluorescence parameter quantum yield (QY) of electron transport of PSII from leaves of hypoxia-stressed plants and further genotyped with 384-SNP Illumina GoldenGate Bead Array. Five quantitative trait loci (QTL) for QY, with a narrow sense heritability of 0.87, were identified on chromosomes 4, 6 and 7H. They had additive effects ranging from 0.74 to 1.35 % with LOD scores from 3 to 12 and explained variance from 6 to 29 %. The major alleles for high QY were from cv. Psaknon; i.e., QY was low if the alleles from cv. Psaknon were not present. Based on leaf necrosis and residual biomass data, the four most interesting QTL may be also in two other populations with completely different progeny, which shows a certain stability of these QTL. The possibility of using marker assistant selection for WLT is discussed, as is possible concurrent improvement of drought tolerance and grain yield.

Plants are known to emit volatile organic compounds (VOC) in response to various biotic or abiotic stresses. Although the VOC emission in the case of insect attacks is well described, there is only little known about the impact of pathogens on plant emission. In the present study, we used a willow-leaf rust system to describe the effects of a biotrophic fungal infection on the VOC emission pattern of willow leaves. We detected that isoprene emissions from rust-infected leaves decreased threefold compared to control. The total monoterpene emissions did not change although a stress-signalling compound (Z)-beta-ocimene showed an increase in infected plants on several days. The infection also increased the emission of sesquiterpenes and lipoxygenase products (LOX) by factors of 175-fold and 10-fold, respectively. The volatile emission signals showed two clear peaks during the experiment. At 6, 7 and 12 days post-infection (dpi), the relative volatile emission signal increased to about sixfold compared to uninfected plants. These time points are directly connected to rust infection since at 6 dpi the first rust pustules appeared on the leaves and at 12 dpi necrosis had developed around several pustules. We present correlations between LOX and sesquiterpene emission signals, which suggest at least two different steps in eliciting the volatile emission.
