Molecular Basis of Neurovirulence of Flury Rabies Virus Vaccine Strains: Importance of the Polymerase and the Glycoprotein R333Q Mutation

Abstract: The molecular mechanisms associated with rabies virus (RV) virulence are not fully understood. In this study, the RV Flury low-egg-passage (LEP) and high-egg-passage (HEP) strains were used as models to explore the attenuation mechanism of RV. The results of our studies confirmed that the R333Q mutation in the glycoprotein (G R333Q ) is crucial for the attenuation of Flury RV in mice. The R333Q mutation is stably maintained in the HEP genome background but not in the LEP genome background during replication in… Show more

“…The ability of foreign proteins to function as new receptor binding proteins has also been demonstrated in other paramyxoviruses (11). RVG mediates both receptor binding and penetration (13,24) and is an important virulence factor for the virus (20,56). The introduction of the RVG gene does not increase the virulence of the NDV vector.…”

“…The modified vaccinia virus strain Ankara expressing the T7 RNA polymerase (64) was grown and titrated in primary chicken embryo fibroblasts. The RV street virus GX/09, isolated from the brain of a dog that died of rabies in Guangxi Province of China in 2009, was propagated and titrated in the brains of adult mice as previously described (56). Recombinant vesicular stomatitis virus (VSV) expressing enhanced green fluorescent protein, rVSV-EGFP was generated and prepared as previously described (38).…”

“…These have been shown to retain residual virulence in some species, and alpacas may have been affected for that reason. 46 Modified live vaccines generated by other methods may present less risk, but this outbreak emphasizes the need for safety testing before using products in large numbers of animals.…”

“…The Ministry of Health has reported 34 cases of human Rabies in 2015 with the maximum number (10) from Kurunegala (MOH Weekly Epidemiological Report), while a total number of 37 cases had been reported in 2012, with 1-5 cases distributed in many districts. (Geue et al, 2008;Tao et al, 2010;Wu et al, 2011). Rabies vaccines produced in compliance with OIE requirements protect against all variants of rabies virus including other phylogroup 1 lyssaviruses (Brookes et al, 2005).…”
