Molecular and functional characterization of erythropoietin of the goldfish (Carassius auratus L.)

Abstract: Erythropoietin is the principal regulator of erythropoiesis and promotes the survival, proliferation and differentiation of erythroid progenitor cells in mammals. In this study we report on the molecular and functional characterization of erythropoietin from the goldfish. Quantitative expression analysis of goldfish epo revealed the highest mRNA levels in heart, followed by brain, liver, spleen and kidney tissues. There was no marked change of epo expression in goldfish primary kidney macrophage cultures, as p… Show more

“…Haematopoiesis in teleosts is similar to that in mammals, as demonstrated by several investigations in zebrafish ( Danio rerio )14, goldfish ( Carassius auratus L.)1516 and carp ( Cyprinus carpio )17. The zebrafish has recently emerged as a powerful genetic model of haematopoiesis1819.…”

“…Despite the importance of erythropoietin hormone in fishes, there is no information available on plasma erythropoietin level and pollutants effect on it in marine fish and just reported some information about other aspects of erythropoietin in freshwater fish such as: effects of hypoxia on tissue EPO levels in freshwater fish (Lai et al, 2006), expression of EPO genes following hypoxia and cadmium exposure in sheepshead minnow (Dangre et al, 2010), molecular and functional characterization of erythropoietin from the goldfish (Katakura et al, 2013). Therefore, to the best of our knowledge, this is the first report on chromium on EPO plasma level in Epaulet grouper as a marine fish.…”

“…Erythropoietin (EPO) is a 30.4 kD glycoprotein hormone that has been identified as a principal regulator of erythropoiesis (Fried, 2009;Katakura et al, 2013). The kidney is the primary erythropoietic organ in teleost fish (Chou et al, 2004) and erythropoietin is produced in proximal tubule cells (Maxwell et al, 1990).…”

“…Moreover, a conserved downstream signaling of erythropoietin receptor has been characterized in zebrafish, homologous to their mammalian counterparts (Paffett-Lugassy et al, 2007;Richmond et al, 2005). Recombinant Epo was shown to support the growth of erythroid progenitor cells in liquid and semisolid media in both fish and amphibian models (Katakura et al, 2013;Nogawa-Kosaka et al, 2011;Stachura et al, 2009Stachura et al, , 2011. Recently, Svoboda et al (2014) demonstrated that recombinant zebrafish Epo stimulates progenitor cells to generate small CFU-E and large BFU-E colonies.…”

“…The gene encoding Epo (epo) has been cloned from many nonmammalian species including pufferfish (Fugu rubripes) (Chou et al, 2004), zebrafish (Danio rerio) (Chu et al, 2007;PaffettLugassy et al, 2007), goldfish (Carassius auratus L.) (Katakura et al, 2013) and African clawed frog (Xenopus laevis) (NogawaKosaka et al, 2010). Studies in non-mammalian systems have shown that epo expression was up-regulated in response to anemia and hypoxia (Nogawa-Kosaka et al, 2010;Paffett-Lugassy et al, 2007).…”
