Molecular and Biological Characterization ofLettuce mosaic virus(LMV) Isolates Reveals a Distinct and Widespread Type of Resistance-Breaking Isolate: LMV-Most

Abstract: Lettuce mosaic virus (LMV) causes an economically important seedborne and aphid-transmitted disease of lettuce and ornamental crops worldwide. The genetic diversity among 73 LMV isolates was examined based on a 216-nucleotide sequence at the variable region encoding the NIb-coat protein junction. Three clusters of LMV isolates were distinguished: LMV-Yar, LMV-Greek, and LMV-RoW. In the latter cluster, two subgroups of isolates, LMV-Common and LMV-Most, accounted for a large proportion of the LMV isolates analy… Show more

“…The infection parameters were compared within rather than across experiments. Reverse transcription-PCR detection of the viral progeny was performed as described previously (31), and the identity of each progeny to the mutant inoculated was assessed by restriction and, in some instances, sequence analysis. Semiquantitative double-antibody sandwich ELISA was performed as described previously (19) after 10ϫ dilution of the plant extracts, so that the relationship between the A 405 and the antigen concentration was linear.…”

“…Two other potyviruses were also used: TuMV, available as an infectious clone of the UK1 isolate (Jenner et al 2000) provided by F. Ponz (Centro de Investigacion y tecnologica del Instituto Nacional de Investigaciones Agraria, Madrid) and propagated on turnip, and LMV-AF199, available in our laboratory (Krause-Sakate et al 2002) and propagated in lettuce (cv. Trocadéro).…”

“…The lettuce (Lactuca sativa L.) is crop originally from Asia, belonging to the Asteraceae family, being the most consumed leafy vegetable in the world (KRAUSE-SAKATE, 2002) and in Brazil (SUTTON et al, 2006). The same can be cultivated during the whole year and in different regions of the country.…”
