“…More recently, nucleotide sequences of the RNAs of BaMMV have been reported: the 3'-terminal half of RNA 1 of two Japanese isolates [16], the RNA1 coat protein gene of an English and German isolate [8,22] and the RNA2 sequence of a German isolate [27]. These sequences show that BaMMV has a similar genetic organization but only a low level of homology to BaYMV, confirming its classification as a distinct bymovirus.…”

“…Total RNA was extracted using the TRIzol reagent (Invitrogen, Paisley, UK) following the manufacturer's instructions. Reverse transcription and PCR were performed using primers designed to regions of homology in RNA‐1 of UK, German and Japanese isolates of BaMMV (Kashiwazaki et al ., 1992; Foulds et al ., 1993; Schlichter et al ., 1993). Forward primer, M3 (5′‐ACA GAG CAC GAG GAG GAA‐3′) and reverse primer, M4 (5′‐GCA TGA GAG ATC TAC CGG‐3′) were used to amplify an 899 bp DNA fragment from the 3′ end of RNA‐1, encompassing the coat protein.…”

“…Capsid protein from isolated virus particles was separated by SDS-PAGE [25], blotted and stained as described previously [36]. Amino acid sequences were analysed with a protein sequencer (477A, Applied Biosystems) with on line PTH amino acid HPLC analysis (120A, Applied Biosystems).…”

“…Among Polymyxa 9raminis-transmitted viruses the complete nucleotide sequence of BaYMV [7, 22, 23, 30-1 and the 3'-half of RNA 1 of barley mild mosaic virus (BaMMV) [24,36] have been published. No sequence data have been reported for WSSMV or WYMV.…”
