Molecular analysis of hormone-regulated petal regeneration in Petunia

Abstract: The petal is an important floral organ of higher plants. To study the mechanism of petal development, the in vitro regeneration system of petals was established in Petunia. High-frequency induction of petals occurred directly from explants on the media containing the combination of N6-benzyladenine (6-BA) and indole-3-acetic acid (IAA). Expression analysis of genes involved in flower development indicated that these genes were classified into three types. ABERRANT LEAF AND FLOWER (ALF) gene was induced during … Show more

“…Plants are able to regenerate the whole body through establishing shoot and root apical meristems in vitro ( Ikeuchi et al 2016 ). Also, different types of floral organs can be induced in vitro, independent of the regeneration of the plant body ( Li et al 2002 , An et al 2004 , Xu et al 2004 , Guan et al 2006 , Wu et al 2008 , Cheng et al 2010 ). In recent years, significant progress has been made towards understanding specification of apical meristems ( Liu et al 2014 , Kareem et al 2015 , Iwase et al 2017 , Meng et al 2017 , Zhang et al 2017 ).…”

“…These results agree with previous studies (Fujii and Shimizu 1990;Tanaka et al 2000; Barakat et al 2010). The regenerated floral tissues can be divided into two types according to whether the tissues are regenerated via callus or directly from explants (Wu et al 2008). In Arabidopsis, stem explants could be regenerated from callus (Guan et al 2006).…”

“…Root explants of St. John's wort regenerated shoots on half strength MS medium with 4.0 mg/l IAA, while only root proliferation occurred with full strength MS medium and the same level of IAA (Goel et al 2009). Also, a high ratio of IAA: cytokinin, was shown to promote shoot formation (Franklin and Dias 2006;Wu et al 2008). A high ratio of auxin to (Table 2).…”

“…It is generally accepted that explant source is an important factor for successful establishment of tissue cultures in many cases. Whole flowers and petals and sepals have been used as explants for plant regeneration in some plant species (Nugent et al 1991;Nakano et al 1994;Wu et al 2008;Mohammadi-Dehcheshmeh et al 2008;Debnath 2005;Gambino et al 2007). To our knowledge, petals of St. John's wort, have not been used as explants for plant regeneration studies.…”
