Hepatocellular carcinoma is the most common type of primary liver cancer in humans. This study aimed to demonstrate anticancer properties of an aqueous extract from Chrysophyllum cainito stem bark (CE) and its underlying mechanisms. Our MTT assay results showed that CE significantly reduced human hepatocellular carcinoma (HepG2) cell viability with the IC50of 100 µg/mL, while human dermal primary fibroblast (HDFa) cells showed less susceptibility in every concentration tested. Determined by Annexin V staining, the proportion of apoptotic HepG2 cells increased in a dose-dependent fashion after 24 hour-exposure of CE. The results from Western blot analysis confirmed that CE reduced procaspase-3, suggesting apoptosis by activating caspase-3 cleavage. Using the DCFH-DA and DiOC6 fluorescent probes, it was found that CE significantly stimulated the generation of reactive oxygen species (ROS) and reduced mitochondrial membrane potential (Δψ m), respectively. According to cell cycle analysis, CE (100 µg/mL) profoundly increased the percentage of cells in the sub-G1 phase, indicating cell apoptosis. These data suggest that CE induces apoptosis and cell death in human hepatocellular carcinoma via generation of intracellular ROS and disruption of Δψm. This is the first demonstration of the anticancer activity with proposed underlying mechanism of CE in liver cancer cells.

This work was designed to estimate the protective effect of Saraca indica L. leaves ethanolic exract against γ‐irradiation induced renal damage in rats. Phytochemical examinations of S. indica L. leaves extract resulted in the separation of three flavanone glycosides: Astilibin (1), Neoastilbin (2), and Eriodictyol‐7‐O‐α‐l‐rhamnopyranoside (3); two flavonols: Quercetin (4) and Quercetin‐3‐O‐α‐l‐arabinopyranosyl‐(1′′′‐6′′)‐O‐β‐D‐galactopyranoside (5) in addition of Gallic acid (6) and methyl gallate (7). Their structures elucidated by chemical evidences and spectroscopic analysis (1 and 2D‐NMR, ‐ESI‐MS, UV). Female rats were used and classified into: control, Ext (200 mg/kg body wt/day orally for 7 days), IRR (8Gy), Ext + IRR, and Sily+IRR groups (received silymarin 50 mg/kg b.wt orally as reference drug). Results showed that S. indica L. leaves extract ameliorated the kidney function tests, hs‐CRP, IL‐1β, ACE, TNF‐α, GSH, and MDA as well as, decreased the histopathological changes of kidney. In conclusion, S. indica L. leaves extract had a renoprotective activity against irradiation induced renal injury due to its flavononid contents.

Introduction:Radiotherapy is an important therapeutic manner of a wide range of tumors. Its side effects on normal cells limit the efficiency of therapy. Aim of the work: To evaluate the protective role of daidzein and/or vitamin C against biochemical, histopathological and ultrastructural changes in male albino rats exposed to gamma radiation (γ-radiation). Materials and Methods: The whole-body exposed to 8 GY γ-radiation. Daidzein was given to rats at a concentration of 63 mg/kg b.wt./day for 7 days before irradiation and 7 days during exposure. Whereas vitamin C was ingested at a concentration of 5 mg/kg b.wt. /day for 7 days before irradiation and 7 days during exposure. Animals were sacrificed on the 1st day post the irradiation dose. Blood samples were collected for biochemical determination of uric acid, urea, and creatinine. Lipid peroxides (TBARS) concentration and the activities of superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx) were also measured. Samples from the kidney were processed for light and electron microscopy. Results: Irradiation induced significant elevation in uric acid, urea, creatinine and TBARS. These changes were accompanied by significant decrease in antioxidant status (SOD, CAT, and GPx). Also, the histological and ultrastructural investigation displayed remarkable changes in kidney tissue. Daidzein and /or vitamin C reduced the kidney damage induced by irradiation; this was showed by declined levels of creatinine, urea and uric acid, in addition to increased CAT, GPx, and SOD activities. The histopathological structure and ultrastructural alternations were also improved. The radioprotection index for the combination of daidzein and vitamin C was found to be more than daidzein or vitamin C separately. Conclusion: Daidzein and vitamin C might modulate radiation damages to kidney by boosting the antioxidant capacity and attenuating oxidative injury.
