Modulation of VP-16 cytotoxicity by carboplatin and 41.8°C hyperthermia

Abstract: These data taken collectively, demonstrate a sequence effect (regarding the aforementioned antineoplastic agents), and provide a framework for future studies directed at the therapeutic optimization of the sequential application of carboplatin, hyperthermia, and VP-16.

“…2). The proton-proton correlations observed in the 1 3.15), together with 1 H-13 C long-range couplings in HMBC spectrum from 2-H, 12-H and 15,17-H to C-13 (d C 136.6) and 14,18-H to C-12 (d C 37.7) established a phenethyl residue. The protons of two singlet methyl groups 32-H (d H 1.42) and 33-H (d H 1.25) were each long-range coupled to an ester carbonyl carbon C-10 (d C 171.6), a quaternary carbon C-11 (d C 54.0) and a ketone carbonyl carbon C-1 (d C 203.7), which in turn were coupled to 2-H.…”

“…GRP78 is a molecular chaperone in the endoplasmic reticulum (ER) induced by ER stress that promotes protein folding and plays an important role as a survival factor in solid tumors by providing resistance to both chemotherapy and hypoglycemic stress [1]. Therefore, specific downregulators of GRP78 transcription can reasonably be expected to become promising drugs in cancer chemotherapy.…”

“…GRP78, which is well-known as a molecular chaperone in the endoplasmic reticulum (ER), also plays an important role as a survival factor in solid tumors, due to its acquisition of a resistant mechanism against both chemotherapy and hypoglycemic stress [1]. Thus, specific down-regulators of GRP78 transcription can reasonably be expected to become promising drugs in cancer chemotherapy [2].…”
