Modulation of TRAIL-Induced Apoptosis by HDAC Inhibitors

Abstract: Triggering apoptosis, the cell's intrinsic death program, is a promising approach for cancer therapy. TNF-related apoptosis-inducing ligand (TRAIL), a member of the TNF superfamily of death inducing ligands, is of special interest for cancer therapy, since TRAIL has been shown to predominantly kill cancer cells, while sparing normal cells. However, since many cancers fail to undergo apoptosis in response to TRAIL treatment, TRAIL-based combination therapies have been developed for cancer-cell specific sensitiz… Show more

“…HDACIs, including TSA, FK228, suberoylanilide hydroxamic acid (SAHA), and LBH589, decrease the expression of Bcl-2, Bcl-xL, and XIAP, and enhance the expression of proapoptotic proteins, such as Bax and Bak, thereby enhancing TRAIL-mediated cytotoxicity in a variety of cancer cells via the amplification of intrinsic as well as extrinsic apoptotic pathways. 90 HDACIs can disrupt mitochondrial membrane permeability and eventually induce the release of proteins, including cytochrome c …”

“…Mechanisms by which different HDAC inhibitors induce apoptosis include activation of both extrinsic and intrinsic apoptotic pathways. HDAC inhibitors have been demonstrated to influence death receptors and their ligands 46 . HDAC inhibitor-induced apoptosis has also been demonstrated to be associated with activation of the intrinsic pathway 45 .…”

“…13 HDACI can revert aberrant epigenetic states in cancer cells that result from aberrant expression or activity of HDACs and histone acetyl transferases. 14 These enzymes determine the acetylation status of histones thereby affecting chromatin topology and gene expression.…”
