Modulation of the Activities of Catalase, Cu-Zn, Mn Superoxide Dismutase, and Glutathione Peroxidase in Adipocyte from Ovariectomised Female Rats with Metabolic Syndrome

Abstract: The aim of this study was to evaluate the association between estrogen removal, antioxidant enzymes, and oxidative stress generated by obesity in a MS female rat model. Thirty two female Wistar rats were divided into 4 groups: Control (C), MS, MS ovariectomized (Ovx), and MS Ovx plus estradiol (E2). MS was induced by administering 30% sucrose to drinking water for 24 weeks. After sacrifice, intra-abdominal fat was dissected; adipocytes were isolated and lipid peroxidation, non-enzymatic antioxidant capacity, a… Show more

“…When analyzing the activity of the antioxidant enzymes, no significant changes in SOD activity were observed in any of the groups, suggesting that this enzyme can maintain its activity regardless of the treatment (HFFD or TB). In contrast, OVX produced a significant decrease in GPx activity, which is associated with the accumulation of H 2 O 2 , which in turn could react with the anion OH-(Fenton reaction) and oxidize cell membrane polyunsaturated fatty acids, increasing the concentration of MDA [41].…”

“…This result is probably because seven days post-OVX or HFFD alone is not sufficient to cause lipoperoxidation and the antioxidant systems are still able to reverse the damage that these conditions could cause. Conversely, when combined, the effects of OVX and HFFD are synergized and able to exceed the antioxidant capacity and, therefore, increase MDA levels [41].…”

“…The mixture was incubated for 3 minutes at 37°C; then 100  μ L of 0.25 mM H 2 O 2 was added to start the reaction and absorbance was monitored for 10 min at 340 nm [11]. Activity is expressed in μ mol NADPH oxidized/min/mg protein.…”
