Abstract: Our data demonstrate that, in vitro, immunoglobulins modulate the activation and cytokine production of peripheral blood lymphocytes from both HD and patients with AD.

“…However, these models have never adequately accounted for the increase of Treg cells after IVIG treatment in psoriasis, skin transplants [18, 19], and experimental autoimmune encephalomyelitis (EAE) [20]. We propose that Tregitopes may be responsible for IVIG-induced tolerance; this hypothesis is corroborated by published reports that polyclonal immunoglobulin therapies induce expansion of Tregs and IL-10 secretion in vivo in animals and humans [21, 22].…”

“…1). It has been demonstrated that CD23 expression on B cells tends to down-regulate following stimulation with IgG1 immunoglobulins and may also be related to antibody-induced B cell modulation [17]. Moreover, CD32, the major Fc-gR expressed by mature B cells, associated with negative B cell receptor (BCR) signalling [18,19] was found to be up-regulated on B lymphocytes at day 15.…”

“…The anti-infl ammatory activity of IVIG is attributed to multiple mechanisms including binding of the IgG Fc domain to Fc-gamma receptors, blockade of the FcRn receptor, blockade of Fas-FasL interactions, or interaction of sialylated Fc with a novel macrophage receptor DC-SIGN (Anthony and Ravetch 2010 ). However, these models have never fully accounted for the observed modulation in T cell phenotype upon co-stimulation with IgG and antigen (Kirschbaum et al 2006 ) and the increase of CD4+/CD25+ Tregs after IVIG treatment in skin transplants (Tha-In et al 2010 ) or experimental autoimmune encephalitis (EAE) (Toubi and Etzioni 2005 ). Others have described the immunosuppressive effects of specifi c IgG-derived peptides subsequently identifi ed as Tregitopes Sharabi et al 2006 ;Hahn et al 2005 ), providing…”
