IntroductionThe first identification of swine-originated influenza A/CA/04/2009 (pH1N1) as the cause of an outbreak of human influenza accelerated efforts to develop vaccines to prevent and control influenza viruses. The current norm in many countries is to prepare influenza vaccines using cell-based or egg-based killed vaccines, but it is difficult to elicit a sufficient immune response using this approach. To improve immune responses, researchers have examined the use of cytokines as vaccine adjuvants, and extensively investigated their functions as chemoattractants of immune cells and boosters of vaccine-mediated protection. Here, we evaluated the effect of Granulocyte-macrophage Colony-Stimulating Factor (GmCSF) as an influenza vaccine adjuvant in BALB/c mice.Method and ResultsFemale BALB/c mice were immunized with killed vaccine together with a murine GmCSF gene delivered by human endogenous retrovirus (HERV) envelope coated baculovirus (1×107 FFU AcHERV-GmCSF, i.m.) and were compared with mice immunized with the killed vaccine alone. On day 14, immunized mice were challenged with 10 median lethal dose of mouse adapted pH1N1 virus. The vaccination together with GmCSF treatment exerted a strong adjuvant effect on humoral and cellular immune responses. In addition, the vaccinated mice together with GmCSF were fully protected against infection by the lethal influenza pH1N1 virus.ConclusionThus, these results indicate that AcHERV-GmCSF is an effective molecular adjuvant that augments immune responses against influenza virus.

Licorice is a medicinal and food plant widely used to treat diseases and produce food additives, because of its unique chemical constituents like polysaccharides, flavones, and saponins. Glycyrrhiza Polysaccharides ( GPS-1 ) are water-soluble neutral polysaccharides extracted from licorice. Currently, GPS-1 is administrated to chickens by gavage every d for 14 d to observe the impact of GPS-1 on the Newcastle disease vaccine. To determine the immunity of these chickens to NDV, blood serum levels of hemagglutinin-inhibition ( HI ) antibody, and immunoglobulins IgA and IgG were measured. Meanwhile, the expression levels of cytokines IL-2, IL-4, IL-17, and IFN-γ were measured to evaluate the degree of immune booster activity. The chickens’ spleen and peripheral blood lymphocytes displayed a significant increase in the proportion of CD4+ and CD8+ T cells after booster treatments with GPS-1. The results indicated that GPS-1 had a significant, dose-dependent, immune-boosting effect which could enhance NDV vaccine immunity in chickens.

La fiebre hemorrágica argentina (FHA) es una enfermedad endemo-epidémica de la República Argentina cuyo agente etiológico es el virus Junín. La población en riesgo de contraer esta enfermedad es de aproximadamente 5 millones de personas que habitan el área endémica. Desde la década de los ‘90 se cuenta con una vacuna para la prevención de la FHA producida de manera local, denominada Candid#1, basada en una cepa atenuada del virus. Sin embargo, dada la naturaleza de la formulación, los elevados costos productivos y los posibles efectos adversos asociados a su administración, es de interés nacional la investigación y el desarrollo de alternativas a esta vacuna. Los baculovirus son virus que infectan insectos, que han sido ampliamente utilizados y estudiados como herramientas de biología molecular por más de 40 años. En la actualidad, existe un sistema de expresión de proteínas heterólogas basado exclusivamente en los baculovirus y células de insectos (denominado BEVS), que constituye una plataforma biotecnológica eucariota, de costos de empleo reducidos y de bajo riesgo biológico. En la presente tesis se exploró la generación de inmunógenos para la prevención de la fiebre hemorrágica argentina mediante el uso de la plataforma BEVS. En el capítulo I se exploró el display superficial de las glicoproteínas de los arenavirus (GPC) en la envoltura de los viriones brotantes de baculovirus recombinantes. En el capítulo II, se buscó generar una línea celular de insecto transgénica, que exprese una enzima esencial para el correcto plegamiento de GPC en el sistema BEVS. Esto se debe a que los anticuerpos neutralizantes responsables de la protección otorgada por Candid#1 reconocen epítopes estructurales de la espícula viral. Luego, en el capítulo III, se evaluó la generación de virus-like particles del virus Junín. Estas partículas son inmunógenos atractivos pues son capaces de exponer los antígenos virales de la misma manera que lo hacen los viriones de los cuales derivan, pero no poseen genoma, por lo cual no son replicativas, y resultan más seguras. Por último se provee de una discusión general de los resultados y la proyección de perspectivas futuras.
