Modulation of gill Na+,K+‐ATPase activity by ammonium ions: Putative coupling of nitrogen excretion and ion uptake in the freshwater shrimp Macrobrachium olfersii

Abstract: The effect of NH 4 þ ions on (Na þ ,K þ )-ATPase hydrolytic activity was examined in a gill microsomal fraction from M. olfersii. In the absence of NH 4 þ ions, K þ ions stimulated ATP hydrolysis, exhibiting cooperative kinetics (n H ¼0.8), to a maximal specific activity of V¼556. þ and K þ ions. These data suggest that at high NH 4 þ ion concentrations, the (Na þ , K þ )-ATPase exposes a new site; the subsequent binding of NH 4 þ ions stimulates ATP hydrolysis to rates higher than those for K þ ions alone. Th… Show more

“…Hsp21 was identified as a differentially expressed gene from a forward SSH cDNA library constructed from gut tissues of P. monodon showing significant increases in gene expression under osmotic stress (Shekhar et al 2013). Ion uptake in crustaceans is mostly attributed to Na + /K + -ATPase and Ca 2+ -ATPase activity (Meyran and Graf 1986;Lima et al 1997;Furriel et al 2004;Masui et al 2005). Several previous studied have found that Hsps could be involved in Na + /K + -ATPase stabilization (Burdon and Cutmore 1982;Deane and Woo 2005;Chatterjee et al 2009).…”

“…+ ions as a substrate, thereby facilitating the active transport of extracellular NH 4 + into the epithelial cell (Furriel et al, 2004;Kurtz and Balaban, 1986;Mallery, 1983;Masui et al, 2002;Nawata et al, 2010a;Skou, 1960;Wall and Koger, 1994). Blockage of the Na + /K + -ATPase may also inhibit ammonia excretion indirectly because of a reduced 'out-to-in' Na + gradient and a consequent reduction in apical acidification via NHE if present.…”

“…Through work on frog skin, planarians, and crustacean and fish gills, a number of cellular mechanisms and transporters for ammonia excretion in freshwater animals have been discovered. In addition to the RhGPs and the Mep/Amts, these include members of the solute carrier family 9 (SLC9, NHEs) (Orlowski and Grinstein, 2004;Zachos et al, 2005); the Na + /K + -ATPase (NKA) (Masui et al, 2002;Furriel et al, 2004;Weihrauch et al, 2012b;Ip et al, 2012); and the V-type H + -ATPase (VA) (Wilson et al, 1994;Shih et al, 2008;Braun et al, 2009;Weihrauch et al, 2009;Wright and Wood, 2009).…”
