Modulation of Gene Expression Profiles by Hyperosmolarity and Insulin

Abstract: Cell hydration changes play a key role in the regulation of cell function and critically affect insulin sensitivity of carbohydrate- and protein metabolism. Here, the modulation of gene expression profiles by hyperosmolarity and insulin was examined in H4IIE rat hepatoma cells by cDNA/oligonucleotiode array-, Northern- and Western blot analysis. Osmosensitive expression of the insulin-like growth factor binding protein Igfbp1, the multidrug resistance protein Mrp5 (Abcc5a) and cyclin D1 (Ccnd1) was established… Show more

“…Potential intermediates may include gluconeogenesis, glycogen breakdown, insulin sensitivity, mitochondrial function, ATP and NADPH generation, and sympathetic activation [69, 1417] (see Figure 1). Reduced mitochondrial ATP production is associated with both hyperosmotic conditions and insulin resistance [15, 65].…”

“…Cell swelling can lower endogenous blood glucose and free amino acid concentrations, by down-regulating gluconeogenesis, glycogenolysis, and proteolysis [69]. At the same time, cell swelling may accelerate glucose clearance by improving insulin sensitivity [8, 9]. Elevated carbohydrate and insulin concentrations prioritize carbohydrate oxidation over fat oxidation, i.e.…”

“…Insulin has been shown to stimulate cellular K ϩ uptake by activation of a Na ϩ /H ϩ exchanger, Na ϩ -K ϩ -2Cl Ϫ cotransport, and Na ϩ K ϩ / ATPase (19 -21). The stimulation of cellular K ϩ uptake was shown to be required for the effect of the hormone on the regulation of metabolism (19 -21) and on gene expression (43). Moreover, the K ϩ uptake may confer support for cell survival (7,8,16,46,48).…”

“…Previous screens identified many hypertonicity-induced genes, including those regulated by NFAT5. Most of those screens employed DNA microarrays, for example (17,20,24,31). Recently, the development of RNA sequencing (RNA-Seq) has provided a means for a more comprehensive profiling of gene expression than previous methods, including DNA microarrays (3).…”
