Modulation of exudate inflammation parameters in rat carrageenan-induced granuloma by modification of exudate iron levels

Abstract: We have used the carrageenan-induced pouch-granuloma in rats to investigate how changes in low-molecular-mass iron chelate levels in the exudate, induced by iron loading (iron-dextran) or chelation (desferrioxamine) influence cellular and systemic inflammatory parameters. In the iron-treated group we observed a rapid decrease in the number of leukocytes and exudate volume; there was also an increase in ferritin iron and low-molecular-mass iron chelates, and on the eighth day a systemic response. In the desferr… Show more

“…After an initial phase of low permeability, a second phase, between 24 and 48 h after induction of inflammation, of increased vascular permeability has been found in experimental inflammatory exudate [16,17]. Although in our study a high cell metabolism, coincident with low local irrigation, may have contributed to retention of CO2 in the exudate during the first hours, after 48h of inflammation an increase in local blood flow may have led to a decrease in the exudate Pc%.…”

“…After day 15 of the experimental feeding period, a granuloma was induced by subcutaneous administration of 6 ml air, followed 24 h later by 4 ml carrageenan (Hispanoland, Barcelona, Spain) 2 % (w/v) in sterile saline, into the dorsum of rats as previously described 24 . Rats were anaesthetized with isofluorane (Abbott Laboratories, Madrid, Spain) 24 h after carrageenan injection.…”

“…A carrageenan granuloma (12) was induced randomly induced in half of the rats of each group at the end of the 16th week of the study diet by an injection of 6 mL of air under the skin on the back, followed by 4 mL of carrageenan 2% (wt/vol) 24 h later. Six days later, nine rats from each subgroup were used for ferrokinetic studies, which lasted 7 d, and the remaining six rats were used for the nonradioactive tests.…”
