Modulation of Endotoxicity of Shigella Generalized Modules for Membrane Antigens (GMMA) by Genetic Lipid A Modifications

Abstract: Background: GMMA from Gram-negative bacteria are an attractive vaccine technology, but lipopolysaccharide (LPS) reactogenicity limits use. Results: Genetic LPS modification resulting in penta-acylation reduced Shigella GMMA reactogenicity to a TLR2-mediated limit. Modifications resulting in palmitoleoylated hexa-acylated LPS triggered higher TLR4-mediated reactogenicity. Conclusion: Use of GMMA as vaccines will likely require LPS penta-acylation.Significance: Understanding the relative contribution of TLR activa… Show more

“…Generalized Modules for Membrane Antigens (GMMA) are OMVs engineered to enhance native OMV formation (by deletion of the gna33 in meningococcus and tolR in Shigella sonnei and Salmonella ), reduce reactogenicity and toxicity (by modifying the acylation pattern of lipid A) and over‐express immunogenic antigens . GMMA contain pathogen‐associated molecular patterns, including toll‐like receptor ligands, which can act as self‐adjuvants in the immune responses they elicit .…”

“…GMMA contain pathogen‐associated molecular patterns, including toll‐like receptor ligands, which can act as self‐adjuvants in the immune responses they elicit . GMMA are highly immunogenic, effective, have low reactogenicity, are safe, and can be manufactured at low cost in a scalable process . The main limitation of GMMA is that it cannot perform human‐like post‐translational modifications (e.g., glycosylation).…”

“…Because humans are more sensitive to LPS than mice, a reaction of human tissues to LPS contained in the vaccine may happen, and this will carefully be examined in future developments. LPS detoxification by deletion of msbB or htrB genes controlling lipid-A acylation could be considered, as previously reported in Shigella [18].…”

“…Several approaches are currently in development to deliver the O-antigen to the immune system, including whole cell attenuated bacteria (6), vaccines in which the O-antigen are chemically-(7) or bio-conjugated to carrier proteins (8), synthetic vaccine conjugates (9), and GMMA based vaccines (10). GMMA are outer membrane exosomes from Gram-negative bacteria, genetically modified to induce hyperblebbing and to reduce the reactogenic potential of lipid A (11, 12). GMMA are easy and inexpensive to produce, and highly immunogenic (10, 1316).…”
