Modulation of differentiation and proliferation in human colon carcinoma cells by transforming growth factor β1 and β2

Abstract: We have previously characterized the diversity of cellular responses to transforming growth factor (TGF)-beta 1 from human colon carcinoma cells. We now show that morphological alteration and part of the growth-inhibitory responses elicited by growth factor (GF) are associated with the secondary effect of the induction of synthesis of extracellular matrix (ECM) glycoproteins. Specifically, morphological alteration is associated with the ECM glycoprotein laminin, and growth inhibition is associated with both la… Show more

“…Transforming growth factor (TGF)-β inhibits the growth and induces differentiation of several lung and other carcinoma cells (Twardzik et al, 1989;Chakrabarty et al, 1990;Schroy et al, 1990). Glucocorticoids synergistically inhibit the growth of monocytoid leukaemia cells in combination with a low concentration of TGF-β (Kanatani et al, 1996).…”

“…Although TGFb can often appear to have different effects in different cell types, its primary effect on colonic epithelial cells is to reduce proliferation and induce differentiation. 30 Mutations of the TGFb receptors are often found in cancer cells with defects in mismatch repair systems. 31 Axin has been shown to regulate the TGFb signalling pathway by acting as an adaptor for Smad3, one of the TGFb effectors ( fig 1B).…”

“…In the parental Moser cells, the expression of three distinct CEA-GLY species of 116,95, and 55 KDa is induced by TGFPl (Chakrabarty et al, 1989(Chakrabarty et al, , 1990aChakrabarty, 1992). In the cloned variant of the Moser cells used in our transfection experiments, TGFPl treatment also induced the cellular expression of CEA and the three CEA-GLY species (Fig.…”
