Modulation of cultured chicken growth plate chondrocytes by transforming growth factor‐β1 and basic fibroblast growth factor

Abstract: Expression of several cellular and matrix proteins which increase significantly during the maturation of growth plate cartilage has been shown to be affected by various endocrine and autocrine factors. In the studies reported here, transforming growth factor-beta (TGF-beta 1) and basic fibroblast growth factor (bFGF) were administered to primary cultures of avian growth plate chondrocytes at pre- or post-confluent stages to study the interplay that occurs between these factors in modulating chondrocytic phenot… Show more

“…When added to postconfluent cells, TGF-␤ 1 was mildly (10-20%) inhibitory to [ 45 Ca] deposition, but when added to preconfluent cells, caused a 2-fold increase. TGF-␤ 1 is unique in its ability to stimulate fibronectin synthesis [Wu et al, 1992]; our current studies show that OP-1 lacks this property.…”

“…We also have studied the effects of TGF-␤ 1 (and bFGF) on chicken GP chondrocytes in serum-free cultures [Wu et al, 1992]. One week of exposure of preconfluent cells to TGF-␤ 1 alone stimulates cell division ( 3 H-thymidine uptake), synthesis of fibronectin and type II and X collagens, and increases ALP activity.…”

“…Recombinant human BMP-2 was a gift from Genetics Institute (Cambridge, MA). 1996) as well as various growth and differentiation factors (Galéra et al, 1992;Wu et al, 1992; Skin fibroblast culture 1993; Böhme et al, 1995;Chen et al, 1995) on cultured chondrocytes from both mammalian and avian sources.…”

“…TGF-b is an important growth factor implicated in regulation of many genes and proteins during chondrogenic maturation [Wu et al, 1992] and is used to induce chondrogenesis in high-density cultures of mesenchymal precursor cells [Lust et al, 1991]. Regulation of FN levels and isoform expression may occur independently of the Smad pathway [Hocevar et al, 1999].…”
