This paper reports the synthesis of haptens and the generation and preliminary evaluation of polyclonal antibodies for the detection of dioxins such as TCDD (2,3,7,8-tetrachlorodibenzo-p-dioxin) by ELISA (enzyme-linked immunosorbent assay). These novel haptens contain unsaturation between the halogenated dibenzo-p-dioxin ring system and the protein to which it is conjugated, presenting a rigid handle structure. The substitution pattern is identical with or similar to that of TCDD (i.e., 2,3,7,8- or 1,2,3,7,8-). Finally, the haptens lack polar groups for hydrogen bonding. In direct binding assays using the new polyclonal antibodies there was excellent recognition of hapten−protein conjugates, including recognition of those hapten conjugates that were not used as immunogens (i.e., assay systems heterologous in hapten structure). These haptens do elicit selective immune responses in rabbits. Their evaluation in an ELISA format demonstrated the usefulness of these haptens for the detection of dioxins. An IC50 of 0.8 ng/well (16 ng/mL) was observed for an unoptimized system that used 2,3,7-trichloro-8-methyldibenzo-p-dioxin as an analytical surrogate standard. A rapid, simple, and reliable competitive immunoassay was developed and validated for measurement of Cd(II) in environmental water samples. This assay employed a monoclonal antibody that recognizes Cd(II)-EDTA complexes as capture reagent and a Cd(II)-EDTA conjugate of horseradish peroxidase as an enzyme label. The assay depended on a competitive binding reaction between the enzyme conjugate and Cd(II)-EDTA complexes, derived from the environmental water sample, for the binding sites of the immobilized antibody. The concentration of Cd(II) in the sample was quantified by the ability of its EDTA complexes to inhibit the binding of the enzyme conjugate to the antibody and, subsequently, color formation in the assay. The assay was specific to Cd(II), with a limit of detection of 0.3 ppb. Ca(II), Mg(II), and Fe(III), the metal ions commonly found in ambient water at relatively high concentrations, did not interfere with the assay. Mean analytical recovery of added Cd(II) was 100.29 +/- 3.60. The precision of the assay was satisfactory; coefficients of variation were 3.6-10.9 and 4.81-10.21% for intra- and interassay precision, respectively. The assay compared favorably with graphite furnace atomic absorption spectroscopy in its ability to accurately measure Cd(II) spiked into water samples from a Louisiana bayou.

Results from polychlorinated biphenyls (PCB) analyses of mussel tissue extracts by immunoassay (PCB RaPID Assay ®) and conventional gas chromatography-electron-capture detection (GC-ECD) are described and compared. Mussels from natural populations with diverse concentrations of PCBs, mussel tissue fortified with technical Aroclor ® 1254 and a certified reference material are included. A strong correlation is reported between "total" PCBs quantified by both techniques (r 2 = 0.95, n = 27). Immunoassay results, however, exhibited lower values compared to GC-ECD, particularly when GC results are corrected for procedural recovery. A reduced antibody response, due to differences in the congener composition between the mussel extracts and Aroclor ® 1254 (used to raise and calibrate the ELISA), provides the most likely explanation for this difference. Non-parametric statistical analyses confirmed that, although differing from Aroclor ® 1254, PCB congener compositions in the mussel extracts most closely resemble that of Aroclor ® 1254. At very high PCB concentrations (>30 g g −1 dry weight), however, ELISA results are statistically different (P < 0.01) from GC-ECD results, which is likely to be related to the solvation capacity of ELISA diluent. Similarity analysis showed high correlations between the most prominent congeners in Aroclor ® 1254 and immunoassay results. This analysis did not, however, identify a specific chlorine substitution pattern to which the immunoassay preferentially responded. Whilst GC-ECD affords the capability to quantify individual congeners of different reactivity and toxicity, the data reported do indicate that immunoassay offers a rapid and inexpensive alternative method for estimation of "total" PCBs at environmentally significant levels. It is, however, necessary to remove extraneous lipids to reduce matrix effects in the immunoassay.
