Modular Evolution of Tn GBS s, a New Family of Integrative and Conjugative Elements Associating Insertion Sequence Transposition, Plasmid Replication, and Conjugation for Their Spreading

Abstract: Integrative and conjugative elements (ICEs) have a major impact on gene flow and genome dynamics in bacteria. The ICEs TnGBS1 and TnGBS2, first identified in Streptococcus agalactiae, use a DDE transposase, unlike most characterized ICEs, which depend on a phage-like integrase for their mobility. Here we identified 56 additional TnGBS-related ICEs by systematic genome analysis. Interestingly, all except one are inserted in streptococcal genomes. Sequence comparison of the proteins conserved among these ICEs de… Show more

“…A more detailed exploration of these results would be useful. Another potential interaction, in this case with RNA polymerase, is suggested by the recent observation that the transposon TnGBS (an ICE from Streptococcus agalactiae) and members of the closely related ISLre2 family insert preferentially 15-17 bp upstream of σA promoters (50,51). Targeting of upstream regions of transcription units has also been extensively documented for certain eukaryotic transposons (121).…”

“…More recent studies (50)(51)(52)(53) have identified ICE with typical DDE Tpases. One group, TnGBS, initially found in Group B Streptococcus, has led to the identification of an entire family of typical insertion sequences, the ISLre2 family (52), whereas another shows a close relationship to IS30 family members (53).…”

“…Upon induction by environmental or intracellular signals, ICEs can excise from the chromosome, usually by site-specific recombination mediated by a tyrosine or a serine recombinase or, in a few atypical cases, by a transposition mechanism mediated by a DDE transposase (11,12,13,14). The resulting circular covalently closed ICE molecule then serves as a substrate for the conjugative machinery that translocates the ICE DNA from the donor to the recipient cell.…”

“…The prolonged extrachromosomal maintenance of ICE6013 in transconjugants strongly suggests that the element is able to replicate before integration, as was shown for ICEBs1, TnGBS, and SXT (44,52,53). We developed a TnSeq-like assay capable of identifying low-frequency chromosomal integrations of ICE6013 in transconjugants that were not detected in their genome assemblies.…”
