Sex pheromones are intraspecific chemical signals that are crucial for mate attraction and discrimination. In Drosophila melanogaster, the predominant hydrocarbons on the cuticle of mature female and male flies are radically different and tend to stimulate or inhibit male courtship, respectively. This sexual difference depends largely upon the number of double bonds (one in males and two in females) added by desaturase enzymes. A mutation was caused by a PGal4 transposon inserted in the desat1 gene that codes for the desaturase crucial for setting these double bonds. Homozygous mutant flies produced 70-90% fewer sex pheromones than control flies, and the pheromonal difference between the sexes was almost abolished. A total of 134 excision alleles were induced by pulling out all or a part of the transposon. The pheromonal profile was generally rescued in excision alleles with a completely or largely removed transposon whereas it remained mutant in alleles with a larger piece of the transposon. Five desat1 transcripts were detected during larval-to-adult development. Their levels were precisely quantified in 24-hr-old adults, a critical period for the production of sex pheromones. Three transcripts significantly varied between control females and males; however, the predominant transcript showed no difference. In mutant flies, the predominant transcript was highly decreased with the two sexually dimorphic transcripts.These two transcripts were also absent in the sibling species D. simulans, which shows no sexually dimorphic hydrocarbons. We also induced a larval-lethal allele that lacked all transcripts and failed to complement the defective hydrocarbon phenotype of mutant alleles.

Voila1 , an enhancer-trap strain in Drosophila melanogaster, expresses GAL4 in most gustatory neurons, both before and after metamorphosis. Voila 1 expression starts at embryonic stage 10. In the periphery, it labels larval gustatory sensilla in the antennomaxillary complex as well as in the pharynx. GAL4 is also expressed in the CNS in a manner that prefigures expression in adult flies. Most Voila 1/1 homozygotes die between second larval instar and early adulthood. Moreover, escaping Voila 1/1 larvae do not show gustatory responses to NaCl and sucrose. The simultaneous rescue of normal larval gustation together with adult viability after removal of the transposable PGAL4 element suggests that both these phenotypes are caused by the same inserted element.

Summary: Based on deletion and complementation mapping and DNA sequencing, a new recessive fully penetrant mutation (DNApol-e pl10R ), causing prolonged larval life and larval/early pupal lethality, is identified as the first mutant allele of the DNApol-e (CG6768) gene of Drosophila melanogaster. A same-sense base pair substitution in exon 1 of the DNApol-e gene is associated with retention of the first intron and significant reduction in DNApol-e transcripts in DNApol-e pl10R homozygotes. Homozygous mutant larvae show small imaginal discs with fewer cells and reduced polyteny in salivary glands, presumably because of the compromised DNA polymerase function following exhaustion of the maternal contribution. Extremely small and rare DNApole pl10R homozygous somatic clones in DNApol-e pl10R /1 imaginal discs confirm their poor mitotic activity. The DNApol-e pl10R homozygotes, like those expressing DNApol-e-RNAi transgene, show high sensitivity to DNA damaging agents. The first mutant allele of the DNApole gene will facilitate functional characterization of this enzyme in the genetically tractable Drosophila model. genesis 50:86-101, 2012.
