Mode of Subacute Sclerosing Panencephalitis (SSPE) Virus Infection in Tissue Culture Cells: III. Neurovirulence of Cell‐Free SSPE Viruses of Niigata‐1, Kitaken‐1, and Biken Strains

Abstract: Cell-free viruses recovered from virus-carrying cultures of the Niigata-1, Kitaken-l, and Biken strains of SSPE virus were examined for neurovirulence. The cell-free viruses were prepared by freezing and thawing or by EDTA treatment of the virus-carrying cultures and inoculated into adult mice intracerebrally. A considerable number of the inoculated mice showed clinical signs about I to 5 weeks after the inoculation. The first symptom was hyperreactivity, which was followed by paresis and myoclonus. All of the… Show more

“…The reason for the poor viral recovery from mice infected with the Biken strain is still unknown. In the tissue culture system, the Biken strain hardly pro ducedcell-free virus (9) and hemadsorption of the infected cells was negative (un publisheddata).…”

“…The cells were grown at 36 C in Eagle's minimum essential medium (MEM) supplemented with 10% bovine serum. The cell-free virus was prepared by freeze-thawing of the infected cultures at the 19th to 21st passage as described previously (9).…”

“…The Biken strain of defective SSPE virus shows neurovirulence in mice and hamsters at low passage levels (Thormar et al, 1978;Ohuchi et al, 1981), but its neurovirulence in hamsters has been found to be decreased after passage for more than 100 generations in HEL cells (S. Ueda, personal communication). In this study, defective SSPE virus which had lost its neurovirulence during passages in HEL cells was transferred to three human neural cell lines.…”

“…Nevertheless, cell-free virus-like particles are needed to study their cell tropism for different cell lines. The freeze-thawing method (43) was unacceptable because our strains could not be reproducibly prepared and the titers were unsatisfactory. Preparation by treating infected cells with cytochalasin D (CD-VLPs) (2) enabled us to infect cell lines and thereby compare the infectivity differences between SSPE virus and MV.…”
