Mobilization of Hematopoietic Stem Cells for Use in Autologous Transplantation

Abstract: Autologous hematopoietic stem cell transplantation (HSCT) is a potentially curative therapeutic approach for various malignant hematologic and lymphoid diseases. Hematopoietic stem cells (HSCs) may be collected from the blood or the bone marrow. HSCs are capable of self-renewal and give rise to progenitor cells, multipotent cells that differentiate and proliferate into the mature cells of the blood and immune system. HSCs and progenitor cells are released from the bone marrow into the peripheral blood through … Show more

“…In addition, the significance of this project is based on the notion that mobilization of HSPCs from the marrow into peripheral blood by G-CSF has become the preferred source of HSPCs for stem cell transplants. 21,22 However, G-CSF fails to mobilize sufficient numbers of stem cells in up to 10% of donors, precluding autologous transplantation in those donors or substantially delaying transplant recovery time. Therefore, additional regimens are needed to increase the number of stem cells in peripheral blood upon mobilization.…”

“…[18][19][20] HSC transplantation is a potentially curative treatment for several hematological malignancies. 21,22 The transplantation procedures are based on HSPC trafficking and their egress from the marrow to the peripheral blood, termed "mobilization," for acquisition of the hematopoietic graft. 21,22 Recent evidence strongly supports a role for eicosanoids in regulation of hematopoiesis.…”

“…The highest value for mean number of CD34 positive stem cells was detected in subgroup IIIa (EPO, 48 hours) then this value was gradually decreased through the 7 days in subgroup IIIb (EPO, 1 week) which could be explained by differentiation of CD34 positive cells and loss of their marker. This coincided with the explanation of other workers who stated that during the differentiation of HSCs, they lose CD34 marker and acquire other biochemical markers specific to a lineage [25] . This could provide an explanation for augmented functional and histological renal recovery after IRI in EPO treated group in this work.…”

“…Um processo padronizado leva à estabilidade dos resultados e diminui as dispersões. A padronização é, também, a base para a capacitação dos profissionais [22][23][24][25] e o uso de procedimentos operacionais padrão evidencia a qualidade da assistência e da competência dos profissionais, priorizando a segurança do paciente 26 .…”
