Mobilization of Bacteroides plasmids by Bacteroides conjugal elements

Abstract: A 4.2-kilobase cryptic Bacteroides plasmid, pB8-51, is found in several colonic Bacteroides species. To determine whether pB8-51 is mobilized by any of the known Bacteroides conjugal elements, we constructed an Escherichia coli-Bacteroides shuttle vector, pVAL-1, which contains pB8-51. We constructed Bacteroides uniforinis 0061 derivatives which carry pVAL-1 and various Bacteroides conjugal elements. The Bacteroides conjugal elements tested were six conjugal tetracycline resistance (Tcr) elements (which appear… Show more

“…That is, pregrowth of strains carrying the elements on low levels of tetracycline (1 ,ug/ml) results in enhancement of the frequency of self-transfer of the element and of the frequency of mobilization of coresident plasmids. One exception is the Tcr Emr element designated Tcr Emr 12256 by Shoemaker et al (16,22). This element exhibits constitutive transfer and mobilization.…”

“…Both types are capable not only of transferring themselves from one Bacteroides strain to another but also of mobilizing coresident plasmids from Bacteroides donors to Bacteroides or Escherichia coli recipients (13,22). Most Tcr elements exhibit tetracycline regulation of self-transfer and mobilization (4,8,13,22,23). That is, pregrowth of strains carrying the elements on low levels of tetracycline (1 ,ug/ml) results in enhancement of the frequency of self-transfer of the element and of the frequency of mobilization of coresident plasmids.…”

“…Escherichia coli strains were grown in Luria broth or Luria broth agar plates. Bacteroides strains were grown in prereduced trypticase-yeast extract-glucose broth or agar (11,17).…”

“…A plasmid containing the largest insert (approximately 9 kbp of NBU2) was chosen for further study. Matings were done as described previously (7,17). Various subclones of the cloned region were tested for mobilization by the IncP plasmids R751 and RP4.…”
