Mn++ BINDING BY PLASMA PROTEINS

Abstract: 54Mn2+ added to human or rabbit plasma in vitro combines selectively with the albumin fraction. At pH 7.0, the Scatchard plot yields a curve that may be resolved for two classes of sites: one, where n1 = 1 and K1 = 2.4 × 104, the other where n2 = 0.5 and K2 = 0.5 × 103.Zn2+, Ni2+ and Co2+ can compete effectively for these sites.

“…Additionally, if Mn is adsorbed on the surface of the MTPLs, this Mn contrast agent would induce a small positive enhancement without heating and partially behave as a liposome marker, independent of membrane destabilization. However, as mentioned previously, the released Mn 21 will rapidly become trapped in the surrounding tissues, [32][33][34] and the doxorubicinMn 21 complexes may act predominantly as an actual drug marker. Therefore, even if the distributions of Mn and doxorubicin are not perfectly identical, the thermotriggered alteration in signal can be used as a macroscopic marker for the release of the substances.…”

“…Second, after release from the liposomes, Mn 21 can rapidly bind albumin or other proteins in the tissue and be retained near the release site. [32][33][34] Third, Mn 21 -albumin or Mn 21 -protein complexes can enhance T 1 relaxivity through the slow molecular moving. [35][36][37] Thus, Mn 21 is an appropriate MRI contrast agent for the design of a thermosensitive, polymer-based, activatable liposomal DDS.…”

“…However, whether Mn can influence the development of DR through other metabolic pathways has yet to be investigated. In plasma, approximately 80% of the oxidized state of Mn 2+ is bound to ALB and globulin and transported to the liver, kidney, small intestine, endocrine glands, pancreas, brain, bone, muscle, and hair 12,28 . ALB is a glycosylated protein synthesized and secreted by hepatocytes, which has anti-oxidative stress and anti-inflammatory 29 .…”

“…1975) and Ni2+ is also bound to albumin in vitro (Rao 1962;Callan & Sunderman 1973;Nandedkar et al 1973). After the administration of 63Ni(C0)4, more 63Ni is present in the erythrocytes than in the serum at short survival intervals (Oskarsson & Tjalve 1979b).…”
