MMP‐2 mediates ethanol‐induced invasion of mammary epithelial cells over‐expressing ErbB2

Abstract: Ethanol is a tumor promoter and may enhance the metastasis of breast cancer. We have previously demonstrated that over-expression of ErbB2 promoted ethanol-mediated invasion of mammary epithelial cells and breast cancer cells. However, the underlying cellular/molecular mechanisms remain unknown. By gelatin zymography, we showed that over-expression of ErbB2 increased the production of matrix metalloproteinase-2 (MMP-2) and MMP-9 in human mammary epithelial cells (HB2). Transient or stable transfection of ErbB2… Show more

“…Conversely, a p38 MAPK inhibitor did not affect P-selectin-induced PI 3-K activation. These data suggest that the activation of PI 3K and p38 MAPK may be independent events in P-selectin-mediated signaling pathway, consistent with previous results [84][85][86][87][88][89].…”

“…In breast cancer cells, heregulin-mediated-PI3K and -p38 MAPK activation has been reported to regulate the activation the matrix metalloproteinase-2 (MMP-2) [84] and MMP-9 [88], proteins that play important roles in cancer cell invasion [85]. Insulin-like growth factor can activate PI3K and p38 MAPK leading to increased breast cancer migration [89].…”

“…As shown in Fig. 2A, the effects may be ascribed to inhibiting MMP2/3 expressions after HIC1 expression, consistent with other group findings that MMP2/3 was responsible for tumor invasion by participating in some signal transduction pathways (19,20). However, HIC1 knockdown in PC3 cells by siRNAs ( Supplementary Fig.…”

“…5,31,32 Zymography is often used to measure MMP-2 level and activity of the conditioned medium, and the result indicates a positive correlation with the cell invasion capacity. 33,34 We found that the Ad-REIC treatment inhibited the level of extracellular MMP-2 in RM-9 cells when determined by zymography, indicating that the suppressed in vitro invasion and in vivo metastasis are attributed to the downregulated MMP-2 activity, at least in part. As both the pro and active forms of MMP-2 were suppressed, it is conceivable that the Ad-REIC treatment could inhibit both MMP synthesis and activity.…”
