MMP-2 is localized to the mitochondria-associated membrane of the heart

Abstract: Matrix metalloproteinase-2 (MMP-2) has been extensively studied in the context of extracellular matrix remodeling but is also localized within cells and can be activated by prooxidants to proteolyze specific intercellular targets. Although there are reports of MMP-2 in mitochondria, a critical source of cellular oxidative stress, these studies did not take into account the presence within their preparations of the mitochondria-associated membrane (MAM), a subdomain of the endoplasmic reticulum (ER). We hypothe… Show more

“…24 Overexpression and activity of MMP-2 and MMP-9 could accelerate the development and rupture of atherosclerotic plaques. 25 In addition to their canonical function in ECM remodeling, researches in the last decade have highlighted new MMP-2 functions, including proteolysis of novel substrates beyond ECM proteins such as troponin I, 26 localization to subcellular organelles such as mitochondria, 27 and proteolysis of susceptible intracellular proteins in those subcellular compartments such as monocyte chemoattractant protein-3. 28 Therefore, inhibition of MMP-2 and MMP-9 is considered as an important target for therapeutic intervention of cardiovascular disease.…”

“…We and others have shown that MMP-2, -7, -8, -9, -12, and -14 increase post-MI. [12, 21-33] In particular, MMP-9 and MMP-12 have been shown to increase at both the gene and protein level, and both pro- and active protein forms are elevated post-MI. [12, 34] Both increase in the infarct at day 1 post-MI and remain elevated through days 5-7 post-MI.…”
