Mixed Lymphocyte Culture Responses of Mice

Abstract: The mixed lymphocyte culture reaction (MLR) ~ through its association with the inheritance of the major histocompatibility antigens has received a great deal of interest in the field of transplantation biology (1). The recent demonstration that specific lymphoid cell types, i.e., 0-antigen-bearing thymusderived lymphocytes (T cells) with allogeneic non-P-antigen-bearing bone marrow-derived lymphocytes ("B" cells), interact to yield a proliferative response, broadens or extends the usefulness of this assay, nam… Show more

“…In previous studies on the BlO.A-BlO,A(2R) combination, Festenstein et al (8) and Alter et al, (1) reported the absence of significant MLC or CML, although Popp did find in vivo lymphocyte transformation in this combination (18). Positive MLC response of BlO.HTG to B10.D2 has previously been reported by Plate (17). Positive CML was found in each direction between the noncongenic strains HTG and BlO,D2 by Nabholz et al , and C3H.0H to (noncongenic) BlO,D2 (15); negative data were obtained for the response of B10.D2 to BlO,D2(RlO3) (S) and a marginal or negative result for the response of ATH to A.SW (15).…”

“…The present study compares MLC and CML responses in congenic strain combinations differing at the entire MHC or with differences restricted specifically to the D region of MHC. Several previous publications have reported significant responses to isolated D-region differences (5,9,12,15,17), whereas others have reported negative results (5,8,12,15); none of these studies has quantitatively compared the strengths of stimulation by whole MHC or D region alone. An effort has therefore been made in the present work to quantitate this comparison by titrating the number of stimulating cells in culture.…”

“…After a further 16-h incubation, 20 fi] 01% trypan blue (v/v) in PBS was added to the pulsed wells and the viability of the cells determined as the percentage of eells exeluding the dye. The supernatant medium was then removed, the cells washed twice with ice-cold DMEM and precipitated with 250 /JI ice-cold 5% TCA (Plate, 1974). The precipitate was washed twice with TCA and solubilized in 0-3 ml 0 25 N NaOH; 0 25 ml of this preparation was mixed with 2 ml scintillation cocktail (Optifluor, Packard) and et/niin incorporated into acid precipitablc material were determined for eaeh well.…”

“…These la antigens are allotypic and appear to be controlled by genes linked to H-2 (8) . The capacity to stimulate in mouse MLR is predominantly expressed by B cells (9) . Studies in mice have progressed quite rapidly due in large part to the availability of inbred strains which allow production of specific antisera detecting I-region cell membrane antigens.…”
