Mitochondrial Targeted Antioxidant SKQ1 Ameliorates Acute Kidney Injury by Inhibiting Ferroptosis

Abstract: Emerging evidence suggests that ferroptosis is highly correlated with the pathogenesis of acute kidney injury (AKI). Ferroptosis, an iron-dependent form of cell death, is manifested by a toxic accumulation of lipid peroxides and ultrastructural changes in mitochondria. We herein investigated the effect of Visomitin (SKQ1), a novel mitochondria-targeting antioxidant, on several AKI models in vivo and in vitro. Our results revealed that SKQ1 treatment greatly reversed renal outcomes in cisplatin, ischemia-reperf… Show more

“…Thus, SkQ1 is able to inhibit ROS, maintain the redox balance in mitochondria and, due to this, reduce the degree of inflammation [34,35]. In addition, SkQ1 has been shown to have beneficial effects in various pathologies [14,36]. Thus, SkQ1 is able to prevent and/or suppress development of all manifestations of accelerated senescence in OXYS rats.…”

“…The ultrastructure of the kidney was imaged by a transmission electron microscope (JEOL JEM-1010, Tokyo, Japan). The percentage of damaged mitochondria was evaluated with ImageJ software as previously described (Song et al 2022 ).…”

“…Hence, this section briefly discusses recent therapeutic approaches targeting ferroptosis. Visomitin, a novel mitochondria-targeting antioxidant, can mitigate mitochondrial ROS production, resulting in decreased levels of lipid peroxidation and ferroptosis, thereby protecting against ischemia- or nephrotoxicity-induced AKI [ 88 ]. Irisin, an exercise-induced hormone known to improve mitochondrial function and reduce ROS production [ 89 ], has been shown to protect against IR injury by upregulating GPX4 in vivo .…”
