Mitochondrial stress management: a dynamic journey

Abstract: Mitochondria undergo continuous challenges in the course of their life, from their generation to their degradation. These challenges include the management of reactive oxygen species, the proper assembly of mitochondrial respiratory complexes and the need to balance potential mutations in the mitochondrial DNA. The detection of damage and the ability to keep it under control is critical to fine-tune mitochondrial function to the organismal energy needs. In this review, we will analyze the multiple mechanisms t… Show more

“…Mitochondrial biogenesis is a complex process, which involve the activation of different signaling pathways that may modify mitochondrial function. Particularly, these pathways act about control of cell metabolism as a physiological response to increased energy demand as well as in regulation of mitochondrial ROS production and detoxification, in the proper mitochondrial respiratory complexes assembly and in the regulation of potential mutations in the mitochondrial DNA [63,64].…”

“…The protein-folding environment in mitochondria is challenged by the complex organelle architecture and the delicate process of assembly of the electron transport chain. In addition, due to their role in energy production, mitochondria undergo continuous additional challenges, such as the management of ROS and the balance in potential mitochondrial DNA mutations [31].…”

“…TAK-242/BTZ treatment increased expression of PINK1, which accumulates at the outer mitochondrial membrane (OMM) and recruits Parkin to initiate mitophagy [47]. Generally, mitochondrial fragmentation precedes mitophagy as mitochondria that are smaller and easier to be engulfed by the autophagosomes [48]. During mitophagy, there is a robust mitochondrial translocation of p62/SQSTM1 which usually binds and targets ubiquitinated proteins to autophagic degradation.…”
