Mitochondrial ryanodine‐sensitive Ca2+ channels of rat liver

Abstract: To examine ryanodine-sensitive Ca channels in mitochondria of rat hepatocytes and their role in energy state of the cells via investigation of the ryanodine effect on mitochondrial membrane potential. Oxygen consumption was measured by polarography using the Clark electrode. The substrates of oxidation such as pyruvate (5mM), α-ketoglutarate (5mM), or succinate (5mM) were used. Oxidative phosphorylation was stimulated by the addition of adenosine diphosphate (200nM). Mitochondrial membrane potential was measur… Show more

“…Preliminary incubation of mitochondria with 10 μM Са 2+ in 0-and Mg-mediums reduced changes of Fluo I t is well known that Ca 2+ activates several key enzymes in the mitochondrial matrix to enhance ATP production [1,2]. So Ca 2+ accumulation in mitochondria is a vital process for functional activity of these organelles [3][4][5][6]. At the same time Moreau et al reveal that the process of Ca 2+ accumulation undergoes complex regulation by Ca 2+ itself [7,8].…”

“…It is known that Ca 2+ concentration elevation in the hepatocytes cytoplasm is accompanied by the rapid transport of these cations into the mitochondrial matrix [15]. This translocation is provided by the presence of such systems as "rapid mode" [15], mitochondrial calcium uniporter [16] and mitochondrial ryanodine receptors [17] in the inner mitochondrial membrane.…”
