Mitochondrial oxidative stress activates COX-2/mPGES-1/PGE2 cascade induced by albumin in renal proximal tubular cells

Abstract: COX-2/mPGES-1/PGE2 cascade is of importance in the pathogenesis of kidney injury. Meanwhile, recent studies documented a detrimental role of mitochondrial oxidative stress in kidney diseases. The present study was undertaken to investigate the role of mitochondrial oxidative stress in albumin-induced activation of COX-2/mPGES-1/PGE2 cascade in renal proximal tubular cells. Following albumin overload in mice, we observed a significant increase of oxidative stress and mitochondrial abnormality determined by tran… Show more

“…The effect disappeared following exposure for an additional 24 h (altogether 48 h) to the same dose. Other groups also reported a decrease in mitochondrial mass in response to Mn-Tbap, although when using a 100-fold higher concentration than ours, and with the drug given in combination with stress induction [85,86]. Forty-eight h of exposure to Mn-Tbap did reduce basal-and ATP-linked OCR along with a significant increase in non-mitochondrial respiration.…”

“…Our present study found that mPGES-1 is activated in the peritoneum of PD patients with UFF, and the expression was also increased in RPMCs treated with high glucose. The results from this study revealed that, in addition to the acute phase of diseases, mPGES-1 is still activated under sustained pathological stimulation, a finding that was consistent with the induction of mPGES-1 in peripheral blood mononuclear cells from hypertensive patients, kidney tissues of the unilateral ureteral obstruction mouse model and renal proximal tubular cells treated with albumin ( 27 29 ). Additionally, in our study, a positive correlation was found between the expression of mPGES-1 in the peritoneum and degree of peritoneal fibrosis.…”

“…An increase in HETEs (8-, 9-, 12- and 15-HETEs, Figure S3B ) and a more depolarized mitochondrial potential with DIC treatment in our study suggest that DIC increases mitochondrial ROS through modifications in oxylipin levels. It has been shown that mitochondrial ROS not only activates PGE synthase, which provides substrate for COX-1 and COX-2 , but also increases COX-2 gene expression [ 47 , 48 ]. Thus, paradoxically, DIC induces the production of COX in multiple ways; the same enzyme that DIC is used to inhibit.…”
