Mitochondrial Mechanisms of Apoptosis and Necroptosis in Liver Diseases

Abstract: In addition to playing a pivotal role in cellular energetics and biosynthesis, mitochondrial components are key operators in the regulation of cell death. In addition to apoptosis, necrosis is a highly relevant form of programmed liver cell death. Differential activation of specific forms of programmed cell death may not only affect the outcome of liver disease but may also provide new opportunities for therapeutic intervention. This review describes the role of mitochondria in cell death and the mechanism tha… Show more

“…Once mitochondrial membrane depolarization occurred and MMP collapsed, cytochrome c would be released from the intermembrane of the mitochondria into the cytoplasm [ 25 ]. KWM treatment induced translocation of cytochrome c , which was proved by examination of the cytochrome c protein level in the cytoplasm and the mitochondria, respectively.…”

“…Mitochondria produce ATP through oxidative phosphorylation, which is necessary for a variety of cellular functions. As mitochondrial metabolism is accompanied by ROS production, the accumulation of damaged mitochondria can activate the mitochondrial apoptotic pathway [31,32] . The mitochondrial apoptotic pathway is one of the main pathways of apoptosis.…”

“…Various pro-apoptotic stimuli can decrease mitochondrial membrane potential and increase mitochondrial membrane permeability. These alterations lead to the release of CytC and other pro-apoptotic factors, accelerate apoptotic vesicles formation, and initiate the caspase cascade reaction, eventually leading to apoptosis [31] . SCI often causes mitochondrial damage and is closely associated with apoptosis.…”

“…Chronic liver disease is caused by viral hepatitis, alcoholic fatty liver disease (AFL), non-alcoholic fatty liver disease (NAFL), drug-induced liver injury (DILI), hepatocellular carcinoma (HCC), and other different etiologies, and the course of disease generally lasts for at least 6 months [ 1 , 2 ]. Some studies have focused on the pathogenesis of chronic liver disease, providing some theoretical support for its prevention and treatment [ 3 ].…”
