Mitochondrial ABC proteins in health and disease

Abstract: ABC transporters represent one of the largest families of membrane proteins that are found in all three phyla of life. Mitochondria comprise up to four ABC systems, ABCB7/ATM1, ABCB10/MDL1, ABCB8 and ABCB6. These half-transporters, which assemble into homodimeric complexes, are involved in a number of key cellular processes, e.g. biogenesis of cytosolic iron-sulfur clusters, heme biosynthesis, iron homeostasis, multidrug resistance, and protection against oxidative stress. Here, we summarize recent advances an… Show more

“…Western blot analysis of mitochondrial protein extracts from MCF-7/ADM cells indicated a specific band at 170 kDa. It was kown that the mitochondrial ABC transporter of dimer, which is regarded as 'half-transporter' was the homologue of the P-gp with half of molecular weight of P-gp (65-94 kDa) (15), and it was not detected in our experiment. This further confirmed the successful extraction of mitochondrial proteins.…”

“…ABCB6 belongs to the family of ATP-binding cassette (ABC) transporters, one of the most abundant families of integral membrane proteins [33]. The ABC transporters couple ATP binding and hydrolysis to the transport of endogenous and xenobiotic substrates across cellular membranes.…”

“…It is localized in the mitochondrial inner membrane, as well as on the cytoplasmic membrane and in the cytoplasm. 30 ABCB7 has an important role in maturation of cytosolic and mitochondrial Fe-S cluster proteins, affecting the function of cytochrome c, NADH dehydrogenase (nicotinamide adenine dinucleotide, the reduced form) and succinate dehydrogenase. It is involved in DNA repair and nucleotide excision repair, in oxidative DNA damage repair, in ribosome biogenesis and tRNA thiomodification.…”
