Mitochondria, oxidative metabolism and cell death in stroke

Abstract: Stroke most commonly results from occlusion of a major artery in the brain and typically leads to the death of all cells within the affected tissue. Mitochondria are centrally involved in the development of this tissue injury due to modifications of their major role in supplying ATP and to changes in their properties that can contribute to the development of apoptotic and necrotic cell death. In animal models of stroke, the limited availability of glucose and oxygen directly impairs oxidative metabolism in sev… Show more

“…Cerebral ischaemia and doxorubicin-induced vascular impairment involve mitochondrial damage [101,102] and preventing mitochondrial damage proved to be a successful novel treatment in these pathologies [103][104][105][106][107][108]. Since SIRT1 has been demonstrated to enhance or restore mitochondrial activity in various tissues [62,87,91,[109][110][111] it is logical to assume that the interference between PARP-2 and SIRT1 expression [62] could be key for the protective phenotype.…”

“…Ischemic stroke is a sudden interruption of blood supply to the brain caused by the blockage of an artery, which may result in brain damage and neurologic dysfunction (3). The suppression of brain damage resulting from ischemic stroke is essential in order to prevent a decrease in the quality of life for patients.…”

“…Mitochondria are the metabolic centers of eukaryotic cells, providing basic energy to numerous types of cellular activities. They are a key factor in determining cell survival and death, and serve an important role in the transduction and expansion of death signals (18). In addition, mitochondria are the primary production site of active oxygen, and exhibit the clearest alterations and damage when cells are suffering from oxidative stress injury.…”
