Objective — Despite the advances in oncology, the prognosis of head and neck squamous cell carcinoma (HNSC) patients remains dismal. In this study, we aimed to determine the relevance of PLCXD2 expression in the tumor microenvironment to the HNSC patient clinicopathological features. Methods — Gene expression analysis and multicolor immunofluorescence histochemistry with HNSC tissuemicroarrays were conducted to examine the relation between PLCXD2 expression and patient outcomes. Additionally, Spearman correlation analysis was used to assess the relationship between PLCXD2 protein expression and tumor immune infiltrating cells (TIICs), as well as immune checkpoints (PD-1, PD-L1, and CTLA-4) in HNSC tissue, while Chi-square test and Cox proportional-hazards models were employed to validate the correlation between PLCXD2 protein levels and clinicopathological characteristics with patient survival. Results — Our findings revealed higher PLCXD2 expression in HNSC tissue compared to control benign tissues. Additionally, we observed a distinct association between the presence of PLCXD2 protein in cancer nests and various TIICs, including CD4+ T cells, CD8+ T cells, dendritic cells, as well as CTLA-4+ cells in HNSC tissues. Furthermore, we demonstrated a correlation between PLCXD2 protein expression in cancer cells and advanced TNM stage, as well as a poorer prognosis. Conclusion — Taken together, this study supports PLCXD2 as an independent prognostic marker and a potentially promising target for immunotherapy in HNSC.

Cancer has become the second leading reason for death in the world. Cell biology states that the bulk of anti‐cancer medications focuses on DNA. A study of how complexes of metals interact with DNA is crucial in creation of the effective chemotherapy drugs. The interactions of DNA with Schiff base and their metal complexes are a thrust area for many researchers, and many papers have reported on them. An amino molecule reacts with carbonyl group to form a variety of chemicals called Schiff bases. Many studies have shown that Schiff bases have compounds that affect biological processes. Due to their potential use as novel reagents for biotechnology and medicine, the research of interactions between transition metal complexes and nucleic acids has grown significantly during the past few decades. It was shown that several metal Schiff base complexes may intercalate DNA. Small molecules can attach themselves to DNA through different ways, such as intercalation and groove binding. The main method to measure nuclease activity was gel electrophoresis, and the ligand alone had much lower activity than the complex. To set the stage for this analysis, a brief overview of Schiff bases and their complexes is provided. Further, this review provides an overview of the DNA interactions brought forth by Schiff base metal complexes. It also draws attention to new developments in the study of Schiff bases and their metal complexes and their prospective use as a bioactive core for DNA cleavage and binding.

Patients with acute promyelocytic leukemia (APL), a subtype of acute myeloid leukemia (AML), often present with severe leucocytosis and thrombocytopenia, which may lead to symptoms that require prompt diagnosis and treatment. Immunophenotyping combined with cytomorphology is a valuable tool for identifying APL patients. We describe the case of a 9-year-old male patient who was diagnosed with APL in July 2022 using immunophenotyping combined with cytomorphology. The patient was treated with chemotherapy and is now in full remission in the maintenance phase. This case demonstrated the importance of timely differential diagnosis using the correct tools, allowing inclusion in the appropriate treatment protocol.
