miRNA profiling of small intestinal neuroendocrine tumors defines novel molecular subtypes and identifies miR-375 as a biomarker of patient survival

Abstract: The aim of this study was to define the miRNA profile of small intestinal neuroendocrine tumors and to search for novel molecular subgroups and prognostic biomarkers. miRNA profiling was conducted on 42 tumors from 37 patients who underwent surgery for small intestinal neuroendocrine tumors. Unsupervised hierarchical clustering analysis of miRNA profiles identified two groups of tumor metastases, denoted cluster M1 and M2. The smaller cluster M1 was associated with shorter overall survival and contained tumors… Show more

“…miR-375 was first identified as a pancreatic islet miRNA regulating insulin secretion (Poy et al, 2004). Although miR-375 expression is low in most cancer types (Yan et al, 2014), high expression of miR-375 is a characteristic of lung and small intestine cancers with neuroendocrine differentiation (Arvidsson et al, 2018;Miller et al, 2016;Nishikawa et al, 2011). In lung cancer, miR-375 is induced by the basic helix-loop-helix transcription factor ASCL1 (Nishikawa et al, 2011).…”

“…The tissue microarray contained 846 tumor biopsies from 412 patients retrieved from patients who underwent surgery for SINET at Sahlgrenska University Hospital in the years 1986 to 2013. Details of the construction of the tissue microarray have previously been described (13). The diagnosis of all tumors was re-validated by staining all tumors for hematoxylin and eosin, synaptophysin and chromogranin A, and reviewed by board-certi ed pathologist (O.N.…”

“…A tissue microarray (TMA) block was assembled as previously described [1]. Briefly, all patients who underwent surgery for SI-NET at Sahlgrenska University Hospital from 1986 to 2013 were included in a tissue microarray.…”
