miR-92a promotes tumor growth of osteosarcoma by targeting PTEN/AKT signaling pathway

Abstract: MicroRNAs (miRNAs) play critical roles in human cancers including osteosarcoma (OS). miR-92a has been found to be a cancer-related miRNA in many cancer types and it is upregulated in OS cell lines. However, the expression and biological function of miR-92a in OS have not been investigated. In this study, we showed that miR-92a expression was increased in OS tissues, and its high expression was correlated with clinical stage, T classification and histological differentiation. Furthermore, patients with high exp… Show more

“…PTEN is known to inhibit the activity of PI3K, which then affects the phosphorylation of Akt (12). The PI3K/PTEN/Akt signal pathway has been demonstrated to play a role in a variety of tumors including hematological neoplasms, liver cancer, intestinal cancer, osteosarcoma, and lymphoma (13)(14)(15)(16). This pathway was recently found to be closely related to chronic inflammatory diseases and autoimmune diseases such as asthma (17).…”

“…In fact, down-regulation of miRNA92 in human plasma is a novel marker for acute leukemia patients [37]. Moreover, miRNA92a was observed to promote tumor growth of osteosarcoma by targeting the PTEN/AKT signaling pathway [38]. Additionally, deregulation of miRNA92a expression has been implicated in hepatocellular carcinoma [39], and, in breast cancer, estrogen receptor expression was reported to be regulated by miRNA92a [40].…”

“…We also observed in COS-7 cells that pSer473 is dispensable for Akt1 signaling and phosphorylation of Thr308 alone was sufficient for maximal Akt1 signal prorogation. These results indicated Thr308 phosphorylation status is a superior biomarker for Akt1 activity [11] and called into question the frequent use of Ser473 phosphorylation as a diagnostic marker for Akt1 activity in cancer patients [12][13][14][15].…”
