miR-577 inhibits glioblastoma tumor growth via the Wnt signaling pathway

Abstract: microRNAs (miRNAs) are commonly altered in glioblastoma. Publicly available algorithms suggest the Wnt pathway is a potential target of miR-577 and the Wnt pathway is commonly altered in glioblastoma. Glioblastoma has not been previously evaluated for miR-577 expression. Glioblastoma tumors and cell lines were evaluated for their expression of miR-577. Cell lines were transfected with miR-577, miR-577-mutant, or control mimics to evaluate the effect of miR-577 expression on cell proliferation in vitro and in a… Show more

“…Glioma usually exhibits a series of malignant features, including extensive, rapid and aggressive progression, resistance to surgery, radiotherapy and chemotherapy, all of which lead to the poor prognosis for most patients 3. Although extensive efforts have been made to improve glioma treatment, average survival time of the patients remains 11‐15 months 4. Consequently, it is imperative to further explore the pathogenesis of glioma and discern potential biomarkers.…”

“…Blocking of the interact complex between Wnt proteins and their coreptors including LRP6 in glioblastoma cells contributes to the anti-tumor effects 53. Knockdown of LRP6 by miRNA and shRNA decrease cell growth in glioblastoma cells 54. Here, we first show that the activation of LRP6/GSK3β/E2F1 in glioblastoma patients is linked with LSH overexpression.…”

“…miR‐515 has also been reported as a tumor suppressor, and similarly, studies have shown that miR‐512 not only inhibits glycolysis but also accelerates apoptosis . So far, there are few reports of a role for miR‐516 or miR‐525 in LUSC, however, there is evidence that they can regulate target genes in other tumors to affect protein expression . miR‐577 affects cell expression by regulating the Wnt signaling pathway, which subsequently affects cell growth and cell viability.…”
