miR‐487b and TRAK2 that form an axis to regulate the aggressiveness of osteosarcoma, are potential therapeutic targets and prognostic biomarkers

Abstract: To investigate the effect of microRNA‐487b (miR‐487b) as well as the underlying mechanism in osteosarcoma (OS). Data downloaded from the Gene Expression Omnibus (GEO) database were used to analyze the expression and prognostic value of miR‐487b/TRAK2. Cell counting kit‐8, colony formation, and transwell assays were performed to investigate the biological functions of miR‐487b and TRAK2. Luciferase reporter assay was applied to confirm the interactions between miR‐487b and TRAK2. miR‐487b was overexpressed in O… Show more

“…Many of these genes have demonstrated associations with the prognosis or progression of gliomas. Notably, EMP3, RGS16, PLAU, TAGLN2, STEAP3, CDHR1, OSMR, SERPINH1, PDPN, CHI3L1, ABCC3, SERPINE1, SOCS1, GPX8, GPNMB, CLEC5A, FOSL2, ICAM1, PRF1 and ITPRIP serve as prognostic markers for gliomas 9,41–52 . Among them, RGS16, CLEC5A and TAGLN2 are key regulatory factors that promote glioma progression by activating the PI3K‐AKT pathway in gliomas 53–56 .…”

“…Moreover, FGF14 and SNX10 have demonstrated tumor suppressive properties in colorectal cancer [ 43 , 44 ]. Conversely, decreased expression of GLB1L2 and TRAK2 has been documented in prostate cancer and osteosarcoma, respectively [ 23 , 45 ]. Hence, these proteins have the potential to serve as good prognostic biomarkers for FMC, especially NmFMC.…”

“…Serum peptidomics has been used to identify late-stage oral melanoma and late-stage oral squamous cell carcinoma in dogs, as well as sarcomeric gene mutation and hypertrophic cardiomyopathy in cat [ 20 , 21 ]. Moreover, profiles of tumor-suppressive peptide biomarkers for ovarian and breast cancers were identified in humans [ 22 , 23 ]. Numerous previous studies have employed proteomics approaches to investigate diseases in cat, including the mucosal proteome in cats with inflammatory bowel disease and alimentary small cell lymphoma, as well as the serum proteome in cats with chronic enteropathies [ 24 , 25 ].…”

“…Work by Chen et al established that miR-487b was significantly downregulated in metastatic tissues compared with matched tumor tissues and that miR-487b can be regarded as a biomarker for early colorectal cancer diagnosis and a criterion to judge metastasis [ 24 ]. Another study by Wang et al illustrated that miR-487b played an oncogenic role in osteosarcoma progression via directly targeting TRAK2, which could advance the development of cancer treatment [ 25 ]. In the present study, considering the dysregulation of miR-487b in the serum of CHB and HBV-related HCC patients, ROC curves were constructed to evaluate the diagnostic value of miR-487b.…”
