Colorectal cancer (CRC) is the third most common cancer and the fourth leading cause of cancer-related mortality worldwide. Aberrant expression of miRNAs play important roles in the development and progression of various types of cancers by modulating oncogenic and tumour-suppressor pathways. Therefore, exploring the functions of microRNAs (miRNAs) that specifically contribute to CRC tumourigenesis and tumour development would greatly aid in obtaining more information on CRC and provide new targets for its diagnosis and treatment. miRNA-411 (miR-411) was previously observed to be aberrantly expressed in multiple human cancers. However, the expression pattern, function and underlying molecular mechanism of miR-411 in CRC remain unclear. Therefore, the present study was performed to detect miR-411 expression, investigate the biological roles of miR-411 and identify its mechanism of action in CRC cells. Here, miR-411 expression was significantly downregulated in human CRC tissues and cell lines, and low levels of miR-411 were correlated with lymph node metastasis, distant metastasis and TNM stage. Resumed expression of miR-411 suppressed cell proliferation and invasion but promoted apoptosis in CRC. Additionally, phosphoinositide-3-kinase regulatory subunit 3 (PIK3R3) was identified as a direct target of miR-411 in CRC. PIK3R3 was upregulated in the CRC tissues and inversely correlated with miR-411 expression. Downregulation of PIK3R3 had tumour-suppressive effects similar to those of miR-411 overexpression in CRC. Moreover, upregulation of PIK3R3 could rescue the tumour-suppressing effects of miR-411 overexpression in CRC cells. More importantly, miR-411 specifically suppressed the activation of the AKT/mTOR signalling pathway in CRC. Therefore, miR-411 functions as a tumour-suppressive miRNA by directly targeting PIK3R3 and indirectly regulating AKT/mTOR signalling pathway. miR-411 may serve as a new therapeutic target for patients with CRC.

Osteosarcoma (OS) is an aggressive bone tumor primarily affecting children and adolescents. The etiology of OS is not fully understood. Thus, there is a great need to obtain a better understanding of OS development and progression. Alterations in miRNA expression contribute to the required molecular alterations for neoplastic initiation and progression. This study is the first to investigate miRNA expression in OS in a large discovery and validation cohort comprising a total of 101 OS samples. We established the signature of altered miRNA expression in OS by profiling the expression level of 752 miRNAs in 23 OS samples using sensitive LNA‐enhanced qPCR assays. The identified miRNA expression changes were correlated with gene expression in the same samples. Furthermore, miRNA expression changes were validated in a second independent cohort consisting of 78 OS samples. Analysis of 752 miRNAs in the discovery cohort led to the identification of 33 deregulated miRNAs in OS. Twenty‐nine miRNAs were validated with statistical significance in the second cohort comprising 78 OS samples. miRNA/mRNA targets were determined, and 361 genes with an inverse expression of the target miRNA were identified. Both the miRNAs and the identified target genes were associated with multiple pathways related to cancer as well as bone cell biology, thereby correlating the deregulated miRNAs with OS tumorigenesis. An analysis of the prognostic value of the 29 miRNAs identified miR‐221/miR‐222 to be significantly associated with time to metastasis in both cohorts. This study contributes to a more profound understanding of OS tumorigenesis, by substantiating the importance of miRNA deregulation. We have identified and validated 29 deregulated miRNAs in the – to our knowledge – largest discovery and validation cohorts used so far for miRNA analyses in OS. Two of the miRNAs showed a promising potential as prognostic biomarkers for the aggressiveness of OS.

Although platinum-based chemotherapies have long been used as standard treatment in ovarian cancer, cisplatin resistance is a major problem that restricts its use. Herein, we investigate the biological function of SLC27A2 and its underlying mechanisms in regulating chemo-resistance in ovarian cancer. The findings show that SLC27A2 down-regulation in primary ovarian cancer tissues correlates with chemo-resistance and poor patient survival in our patient cohort. Significantly, we demonstrate that up-regulation of SLC27A2 by lentivirus-mediated p-SLC27A2 sensitizes ovarian cancer cells to cisplatin in vitro and in vivo via apoptosis. Mechanistic investigation reveals that miR-411 is the most strikingly over-expressed gene in response to ectopic expression of SLC27A2, but under-expressed in recurrent ovarian cancer tissues. Lower miR-411 expression contributes to ovarian cancer chemo-resistance in vitro and in vivo. Furthermore, SLC27A2 directly binds specific sites in the miR-411 promoter region and promoter activity decreases after mutation of putative SLC27A2-binding sites. This indicates that SLC27A2 is required for the transcriptional induction of miR-411. The luciferase assays also confirm that miR-411 directly targets ABCG2 in ovarian cancer, and overall findings establish the SLC27A2-miR-411-ABCG2 pathway in the regulation of ovarian cancer chemo-resistance with potential therapeutic applications.
