miR-340 alleviates chemoresistance of osteosarcoma cells by targeting ZEB1

Abstract: Chemoresistance during treatment of osteosarcoma (OS) is attracting more and more attention as the main clinical obstacle. The purpose of this study was to elucidate the role of miR-340 in chemoresistance of OS. Plasmid construction and transfection, miRNA arrays, PCR analyses, and western blot analysis, as well as MTT, apoptosis, and luciferase assays were carried out in MG-63 cells and MG-63/cisplatin (DDP)-resistant cells. The results showed that miR-340 was downregulated in OS tissues and drug-resistant OS… Show more

“…This finding suggests that ZEB1 takes part in the miR-708-5p-mediated suppress of the migration and invasion of OS cells. ZEB1 is highly expressed in OS tissues when compared to normal tissue (18,40). Moreover, the protein level of ZEB1 was found to be upregulated in OS MG63 and SaOS-2 cells unlike that in the hMSCs and HS-5 cells.…”

“…MIAT promotes ZEB1 expression in osteosarcoma. ZEB1 is important for the proliferation and invasion of osteosarcoma cells (19). Thus, it was investigated whether MIAT affects ZEB1 expression in osteosarcoma cells.…”

“…[18][19][20] The administration of synthetic miR-590-3p lipid formulations immediately after MI in mice results in marked reduction of infarct size and persistent recovery of cardiac function. 18 Besides, we previously predicted that zinc finger E-box binding homeobox 1 (ZEB1) gene, which is a transcription factor and is known for the proliferation and invasion of various cells 21,22 including human cardiomyocytes, 10 is the potential target of miR-590-3p.…”
