miR-29b inhibits the progression of esophageal squamous cell carcinoma by targeting MMP-2

Abstract: Accumulating evidence has shown that microRNAs (miRNAs) are aberrantly expressed in human esophageal cancer and crucial to tumorigenesis. Herein, we identified the role of miR-29b in esophageal squamous cell carcinoma (ESCC) development in vitro and in vivo. MiR-29b expression was investigated in thirty cases of ESCC samples as well as four ESCC cell lines by real-time PCR. Transwell assays were conducted to explore the effects of miR-29b on the invasion of human ESCC cell lines. The impact of overexpression o… Show more

“…Yuan et al have found that miR-29b as a suppressor could inhibit the growth of colorectal cancer cells by binding IGIF1, an activator of PI3K/Akt signaling [ 16 ]. In 2015, Qi et al proved that the overexpression of miR-29b significantly reduced the protein of MMP-2 which suppressed the cell invasion of esophageal squamous cell carcinoma [ 17 ]. Meanwhile, the miR-29b was suggested to inhibit the migration and invasion of nasopharyngeal carcinoma cell lines in vitro [ 18 ].…”

“…However, it is worth to note that miR-29b acts as a tumour suppressor in other cancer tissues. Indeed, mir-29b inhibits the progression of esophageal squamous cell carcinoma by targeting MMP-2 42 . In breast cancer, miR-29b expression is negatively associated with the HER2 sub-type, and its overexpression inhibits breast cancer cell proliferation and induces apoptosis mainly downregulating STAT3 protein levels 43 .…”

“…In addition, inhibition of CCAT2 decreased the expression levels of b-catenin/WISP1 downstream targets VEGF-A, MMP2, and ICAM-1, which play vital roles in cancer angiogenesis, migration, and invasion. [38][39][40] Targeting the CCAT2/b-catenin/WISP1 axis may inhibit metastasis and increase radiotherapy sensitivity in ESCC patients by reversing the EMT-associated progression. Moreover, patients with high expression levels of both CCAT2 and WISP1 had the worst prognosis, suggesting that CCAT2 combined with WISP1 may be prognostic biomarkers for ESCC.…”
