MiR-203 Interplays with Polycomb Repressive Complexes to Regulate the Proliferation of Neural Stem/Progenitor Cells

Abstract: SummaryThe polycomb repressive complexes 1 (PRC1) and 2 (PRC2) are two distinct polycomb group (PcG) proteins that maintain the stable silencing of specific sets of genes through chromatin modifications. Although the PRC2 component EZH2 has been known as an epigenetic regulator in promoting the proliferation of neural stem/progenitor cells (NSPCs), the regulatory network that controls this process remains largely unknown. Here we show that miR-203 is repressed by EZH2 in both embryonic and adult NSPCs. MiR-203… Show more

“…miR-203 located on human chromosome 14q32 is a putative tumor suppressor gene. RE1 silencing transcriptional factor (REST), EZH2 and SNAI1/2 are upstream transcriptional repressors of miR-203 [ 84 , 85 , 86 , 87 ].…”

“…Deletion of p63 in mice results in a dramatic loss of all keratinocytes and loss of stratified epithelia, probably due to a premature proliferative exhaustion of the stem and transient amplifying cells [ 89 ]. On the other hand, the EZH2-miR-203-BMI1 regulatory axis functions for the proliferation of neural stem/progenitor cells [ 87 ]. Overexpression of miR-203 suppresses the expression of BMI1 and colony formation in esophageal squamous cell carcinoma cell line [ 90 ].…”

“…Interestingly, miR-203, a microRNA we have recently found to inhibit proliferation via Bmil repression in NSPCs [31], was under regulation of bivalent promoters and dis-played an increased histone Kcr level at its proximal promoter regions, as well as upregulation of expression after crotonate treatment in NSPCs (Fig 5E-5G and S7A-C Fig). In consistent with our previous findings, a reduction of both mRNA and protein levels of Bmi1 was observed concordant with up-regulation of miR-203 (Fig 5H and 5I).…”

“…Additionally, other miRNAs targeting epigenetic regulation have been found to play a role in proliferation. Differentiation induced downregulation of miR-203 occurs in NSCs and negatively regulates proliferation through targeting Bmi1 mRNA which is part of a well-studied repressive epigenetic complex, the Polycomb Repressive Complex (PRC) ( Liu P.P. et al, 2017 ).…”
