miR‐19b‐3p induces cell proliferation and reduces heterochromatin‐mediated senescence through PLZF in goat male germline stem cells

Abstract: Promyelocytic leukemia zinc finger PLZF, known as ZBTB16 or ZFP145 is a critical zinc finger protein of male germline stem cells (mGSCs), it's an essential transcriptional factor for goat testis development and spermatogenesis. Loss of PLZF results in progressive depletion of SSCs after the first wave of spermatogenesis leading to eventual spermatogenic arrest, apparently the result of a shift in the balance in SSC fate away from self-renewal and toward differentiation. Cumulating evidences have demonstrated t… Show more

“…In particular, miR-214 and miR-125b have been found to operate as key mediators of HF maintenance in the telogen stage through their suppressive roles in HFSCs proliferation and differentiation [11]. Similarly, miR-19b-3p was confirmed to be associated with an enhanced proliferation of goat male germline stem cells [12]. Furthermore, miR-19b-3p was demonstrated to target mitogen-activated protein kinase 1 (MAPK1) in embryonic fibroblasts from the great tit under hypoxic conditions [13].…”

“…In our research, the expression profile of miRNAs had obvious growth stage specificity, in which miRNAs associated with testis development and spermatogenesis were expressed at specific growth stages. For example, miR-106b, miR-19b, miR-27a-3p, miR-27b, miR-31, and miR-335 were expressed specifically on the juvenile stage ( 61 66 ); miR-449a was expressed specifically on the adolescence stage ( 32 ); miR-296-3p was expressed specifically on the adult stage ( 67 ). The results suggested that the growth period-specific miRNAs have an important influence on testis development and spermatogenesis.…”

“…However, different results have also been observed in other kinds of studies. For instance, miR-19b was shown to negatively regulate PLZF expression in goat mGSCs and induce goat mGSC proliferation [22]. Kong also pointed out that miR-19 promoted skeletal muscle cell differentiation cooperatively with miR-17 [23].…”
