MiR-185-3p mimic promotes the chemosensitivity of CRC cells via AQP5

Abstract: Background: Studies showed that microRNAs (miRNAs) are important regulators in drug resistance. The current study investigated the role of miR-185-3p and its predicted target gene AQP5 in 5-FU-insensitive colorectal cancer (CRC) cells. Methods: Quantitative real-time polymerase chain reaction (qRT-PCR) and Spearman's correlation analysis were conducted to determine the correlation of expression levels of miR-185-3p and AQP5 from CRC tissues. HCT-116 and HCT-8 cells were treated by gradient concentration of 5-F… Show more

“…MiRNA plays a momentous role in the progression of CRC by up-regulating oncogenes or down-regulating oncogenes [33]. For example, the latest research found that miR-185-3p is down-regulated in CRC, and upregulating miR-185-3p can enhance the chemotherapy sensitivity of CRC cells by targeting AQP5 [34]. Ma et al found that knocking down LINC02163 attenuated CRC cell proliferation and metastasis through the miR-511-3p/AKT3 axis [35].…”

“…MiR-185-3p, also upregulated in the mutation group, is proven a tumor suppressor in colorectal cancer (CRC) 34 , the overexpression of which could enhance the chemosensitivity of CRC patients. Therefore, it is considered a potential target for the treatment of 5-FU-insensitive CRC 35 .…”

“…The primers are listed in Table 1 . U6 ( Zhou C. et al, 2020 ) and β-actin ( Li L. et al, 2018 ) act as endogenous controls. Samples were analyzed with the comparative CT method, where fold change was calculated from the ΔΔCT value with the formula 2 –ΔΔ Ct .…”

“…By repressing the level of neurofibromatosis 1, the upregulated expression of miR-641 promotes resistance to ER in non-small-cell lung cancer cells (Chen et al, 2018). Previous studies indicated that miR-185-3p can promote the antitumor effect of other antitumor drugs (Uhr et al, 2019;Zhou C. et al, 2020). However, it is not clear whether miR-185-3p affects the ER resistance in lung cancer.…”
