Biological function of microRNA-20c-3p (miRNA-520c-3p) in the progression of nasopharyngeal carcinoma (NPC) and the potential mechanism were investigated. Relative level of miRNA-520c-3p in NPC tissues and adjacent normal tissues was determined by quantitative real-time polymerase chain reaction (qRT-PCR). Particularly, miRNA-520c-3p level in NPC with different tumor stages and tumor sizes was examined. Subsequently, miRNA-520c-3p level in nasopharyngeal epithelial cells and NPC cells was detected. The potential influence of miRNA-520c-3p on the proliferative ability and cell cycle progression of NPC cells were evaluated through cell counting kit-8 (CCK-8) and flow cytometry. The target gene of miRNA-520c-3p was verified by dual-luciferase reporter gene assay. Regulatory role of miRNA-520c-3p/RAB22A in the malignant progression of NPC was identified. miRNA-520c-3p was downregulated in NPC tissues and cell lines. Its level was lower in NPC with worse tumor grade and larger tumor size. Overexpression of miRNA-520c-3p suppressed the proliferative ability and arrested cell cycle in G0/G1 phase. RAB22A was confirmed to be the downstream target of miRNA-520c-3p. In NPC tissues and cell lines, RAB22A remained in higher abundance relative to controls. Overexpression of RAB22A reversed the inhibitory effects of overexpressed miRNA-520c-3p on proliferative ability and cell cycle progression of NPC cells. miRNA-520c-3p is downregulated in NPC, which accelerates the malignant progression of NPC by targeting RAB22A.

Nasopharyngeal carcinoma (NPC) is a disease that is highly associated with the latent infection of Epstein–Barr virus. The absence of obvious clinical signs at the early stage of the disease has made early diagnosis practically impossible, thereby promoting the establishment and progression of the disease. To enhance the stride for a reliable and less invasive tool for the diagnosis and prognosis of NPC, we synopsize biomarkers belonging to the two most implicated biological domains (oncogenes and tumor suppressors) in NPC disease. Since no single biomarker is sufficient for diagnosis and prognosis, coupled with the fact that the known established methods such as methylation-specific polymerase chain reaction (PCR), multiplex methylation-specific PCR, microarray assays, etc., can only accommodate a few biomarkers, we propose a 10-biomarker panel (KIT, LMP1, PIKC3A, miR-141, and miR-18a/b (oncogenic) and p16, RASSF1A, DAP-kinase, miR-9, and miR-26a (tumor suppressors)) based on their diagnostic and prognostic values. This marker set could be explored in a multilevel or single unified assay for the diagnosis and prognosis of NPC. If carefully harnessed and standardized, it is hoped that the proposed marker set would help transform the diagnostic and prognostic realm of NPC, and ultimately, help prevent the life-threatening late-stage NPC disease.

Nasopharyngeal carcinoma (NPC) is a rare cancer in the Western hemisphere (1 case per 100,000 individuals). Conversely there are some areas, such as Southern China, Southeast Asia (Vietnam), the Mediterranean basin and Alaska, where morbidity reaches 80 per 100,000 individuals. Detecting cervical lymph node metastasis is important because it helps recommend the radiation dose and the chemotherapy strategy. We compared the diagnostic value of hematoxylin-eosin (HE) staining and a immunohistochemistry technique to determine the pathological features of tumors, in 53 samples of cervical lymphadenopathy from 30 patients with NPC. With HE, metastasis was detected in 41 of 53 samples with NPC and cervical lymphopathy. In the remaining 12 slides, the HE staining did not show signs of metastasis. Staining with immunohistochemistry revealted that 10 of 12 "negative" diagnoses based on HE staining, were actually metastatic. This means that 20% of the patients would not receive correct treatment based on HE staining alone.
