miR‐124 inhibits cell proliferation in gastric cancer through down‐regulation of SPHK1

Abstract: SPHK1 expression is elevated in gastric cancer and is associated with shorter survival times for patients. However, the molecular mechanism of SPHK1 up-regulation in gastric cancer remains unclear. In the present study, we report that miR-124 down-regulated SPHK1 expression by directly targeting its 3'-untranslated region (3'-UTR) and that miR-124 expression was inversely correlated with SPHK1 expression in gastric cancer samples. Furthermore, we demonstrated that, similar to the effect of silencing SPHK1, up-… Show more

“…The current authors previously reported that the miR-124 expression was down-regulated in cutaneous SCC (14), which results in the overexpression of ERK as a target molecule and subsequent cell proliferation. miR-124 was also reported to be involved in the carcinogenesis of various cancers such as glioblastoma, gastric cancer, hepatocellular cancer, breast cancer, and prostate cancer (15)(16)(17)(18)(19), indicating that miR-124 is a key miRNA in carcinogenesis.…”

“…Weng et al suggested that honokiol attenuates the severity of acute pancreatitis and lung injury through suppression of apoptosis and caspase-3 activity (44). Honokiol also inhibits the activation of caspase-3 and caspase-9 in H 2 O 2 -induced apoptosis in human lens epithelial cells (45).…”

“…For instance, miR-124 inhibits migration and invasion of ovarian cancer cells by down-regulating SphK1 and reduces gastric cancer cell proliferation also by targeting SphK1 [26,27]. miR-124, which regulates NFATc1, CAMTA1 (calmodulin-binding transcription activator 1) and PTBP1 (polypyrimidine tract-binding protein 1) to reduce nuclear factor of activated T cells (NFAT)-dependent transcription, is down-regulated in human PASMCs and mouse lungs under hypoxic conditions [28].…”
